| The objectives were to evaluate the expression of AR in different tissues and cellular localization in testis, epididymidis.4-month-old hybrid lambs (Dorper x Han sheep) evaluate the expression of AR in different tissues and cellular localization in testis, epididymidis.2,4,6,12-month-old hybrid lambs evaluate the expression in testis, epididymidis. The expression of AR in various tissues such as heart, liver, spleen, testis, epididymidis and so on, and the Cellular localization of gonad of testis and epididymidis. Total RNA was isolated from various tissues, and mRNA expression was detected by real-time fluorescent quantitative PCR. Cellular localization of AR in testes and epididymidis were examined by immunohistochemistry, respectively, qualitative investigation of the AR protein expression by Western blotting. The main findings are as follows:1. The partial length cDNA of sheep AR gene was obtained firstly and submitted to the NCBI. The result of alignment of nucleic acid sequence of sheep AR with AR in NCBI (ID:NC019484) showed that homology of sheep and AR in NCBI is the highest with the similarity,100%respectively. It indicates that the part sequence of sheep AR gene can be Real-time PCR analysis.2. AR was expressed in many tissues. The quantitative real-time PCR results showed the rank order of AR mRNA expression was testes>heart>abdominal>EpB>brain>small intestine>hindquarters>Longissimus dorsi (LD) muscle> Spleen>EpH> Kidney>intestinum crassum> Liver>Lung>Forequarters>EpT, and the Western blotting results showed the rank order of AR protein expression was testes>heart>abdominal>brain>cerebellum>small intestine>hindquarters>Longissimus dorsi (LD) muscle>Spleen>Forequarters>Liver>Lung>abomasums. It tells us AR plays an important role in the development and maturation of sperm and the gonads and the protection in the heart.3. This test determines the AR of sheep testicles were expressed, in different periods; it’s hardly expressed in epididymis. It is a large number expression of4-month-old testis. The expression of AR in pseudostratified ciliated columnar epithelium and interstitial tissuein EpH, EpB, EpT showed a weak positive signal. There was a positive expression of AR in interstitial tissue, spermatogenous cells and myoid cells of sheep testes. The expression levels of testicular much higher than the epididymis expression in different physiological stages. AR plays an important role in the occurrence of the animal behavior and maintenance of male secondary sex characteristics. |
