Lactuca sativa attributes lettuce in the composite family and it is used from leaves. It is also called Ezai Cai or Wozai Cai, which are annual and two years’for maturity vegetables. They are two of the most popular vegetables in China because of their high value in nutrition. As the plantation scale have been increasing for recent years, as well as the change of ecological environment, the growing of Lactuca sativa are heavily infected by virus, which leads to a huge damage in economics within relative industry. Researcher found that the Lactuca sativa in Shanxi Agricultural University’s test fields suffered heavily dwarf and shrinking during the year of2011and2012. Systematic test were conducted by using inoculation, dsRNA solation technique, PCR amplification (SIA), RT-PCR and some other methods. The results showed that:(1) Inoculation test showed that virus probably infected the lactuca sativa.(2) Base on the RNA (dsRNA) of virus, as well as the SIA method, which using the arbitrary primers, the test showed that the latuca sativa in Shanxi Agricultural University test firms is infected by Cucumber mosaic virus (CMV) and Potato virus S (PVS), and they are named CMV-SC and PVS-SC respectively.(3) In order to know the molecular evolution of CMV-SC and PVS-SC, researchers did clone of whole genome of these two viruses. The result presented CMV-SC has the highest homology with SXCH, sub-group of IB from Taigu, Shanxi Province. Researchers attributed CMV-SC to the sub-group IB. The PVS-SC has the highest homology with WaDef from The United States.(4) To make sure of the variation of amino acid, researchers compared the amino acid from CMV-SC and PVS-SC with those of SXCH and WaDef. They found that la protein, represented by SC-RNA1has9different amino acid sites. The different amino acid sites of2a and2b represented by SC-RNA2are9and11respectively. The number in MP and CP protein, which are represented by SC-RNA3, are2and10respectively. At the same time,223KDa helicase,25K,12K,7K, CP protein, which are all represented by PVS-SC are found to have5,7,6,4,3different amino acid sites respectively.
