The brown planthopper (BPH), Nilaparvata lugens (Stal), is one of the most devastating rice insect pests in China and many other Asian countries. Chemical control is the main method to protect rice against the BPH. However, the BPH shows resistance to chemical agents because of widely use. RNAi is a powerful tool for gene function determination and gene silence. It is studied to control cancers and viral disease. And it also can be an alternitive to control insect pests.In this study, the4target genes including two sucrase-like genes (named145,282) and two sugar transporter-like genes (named303,246) were chosen. The dsRNA under the control of a Zea may polyubiquitin-1promoter (ZmUbi) was expressed. The the glyphosate resistance gene GlOevo(EPSPS) was applied as the selection marker. The transgenic rice expressing different dsRNA were obtained by agrobacterium-mediated transformation method.BPH bioassays showed that the To generation of transgenic rice had no insecticidal activity to BPH. A slight decrease on the number of BPH was due to escape or death from mechanical damage. The seeds of seven rice events chosen from To generation of transgenic rices above-mentioned were sowed. Glyphosate diluted200folders was sprayed to select positive transgenic plants in the five-leaf stage. Northern blot analysis showed high expression of dsRNA in some transgenic rice lines. Three rice events with a high dsRNA expression were chosen for BPH bioassay. No BPH resistance was detected in those transgenic rices. The results indicate that dsRNA could be stably expressed in transgenic plants, but transgenic rice did not show resistance to BPH.
