Rice black streaked dwarf virus(RBSDV) is a viral disease of rice, with non-scheduled, mobile and explosive outbreaks, causing a great threat to rice production in China. Using sequence information from the RBSDV, we made RNA interference (RNAi) constructs and transferred them into the sensitive variety wulingjingl to generate transgenic plants. Then, we performed PCR and qRT-PCR to identify transgenic positive plants, following by inoculation with RBSDV through the plant hopper to screen transgenic lines displaying high resistance against this virus. The main results are as follows:1.We identified T1 generation of the RNAi transgenic plants, and detected their expression level from transgenic positive lines.2.We generated T3 pure lines of pANDA-S8, pANDA-S9 and pANDA-S8&S9.3.We used high expression of transgenic T3 pure lines from pANDA-S8, pANDA-S9 and pANDA-S8&S9 to detect their resistance against RBSDV in the field by inoculation of the virus through the plant hopper. The results showed that compared with non-transgenic plants, these transgenic plants displayed enhanced resistance to the virus, and the chimeric transgenic plants were most resistant to the virus.
