| It is reported that the Fols Paulowniae maintains the effects of anti-inflammation,anti-bacteria, anti-tussive and hypotensive. Up to now, researched mainly focus on thechemical feathers of Fols Paulownia, as well as its antibacterial and anti-inflammatory effects,however the biological activities on immune system are not clear. In this present study, theeffects of flavonoids Fols Paulownia composition on immune system of mice wereinvestigated.Object: The aim of this study is investigating the immune effects on mice both in vivoand in vitro.Methods:The optimal extraction temperature, time, solid-liquid ratio and extraction agentconcentration were determined by orthogonal design, then a standard curve was plotted byRutin, which served as a standard solution, finally, the optimum extraction processwasobtained and the yield of Fols Paulownia flavonoids was determined. The effects ofFlavonoids Fols Paulownia on lymphocyte proliferation were determined by the blastisation of spleen lymphocyte (MTT assay). The effects of Flavonoids Fols Paulownia on humoralimmunity of mice were determined by hemolysin assay and hemolytic plaque assay afterconcentrations (1000ã€500ã€250ã€125ã€62.5mgï¼kg) of Fols Paulownia flavonoids treatment.Results:1. The study indicate that the extraction conditions of flavonoids of FolsPaulownia by orthogonal optimum are90℃(temperature),2h(time),1:25(Solid-liquid ratio),60%(concentration of Ethanol concentration) and the rate of production is4.792%.2.The effects of flavonoids of Fols Paulownia on proliferation of spleen lymphocyte ofmice induced by ConA show that there are significant differences comparing the drug groupwith kinds of concentrations with the negative group of saline as well as the positive group oflevamisole(P<0.05). The result of flavonoids of Fols Paulownia on proliferation of spleenlymphocyte of mice induced by LPS indicate that it is significantly different when comparingthe negative control group of saline with the positive control group of the drug namedlevamisole(P<0.05), however it is not significantly different when comparing the group withkinds of concentration drugs with the negative control group of saline as well as the positivecontrol group of the drug named levamisole(P>0.05).3.The effects of Fols Paulownia flavonoids on humoral immune function, hemolysin andhemolytic exhibited a concentration dependent manner that it is significantly different when comparing the negative control group with the positive control group as well as the druggroup(P<0.05)and the most remarkable concentration is1000mg/kg.Conclusion:1. The optimum extraction condition of Royal Fols Paulownia flavones areas followed: temperature is90℃, time is2h, solid-liquid ratio is1:25, and ethanolconcentration is60%, then the product yield is4.792%.2. Royal Fols Paulownia flavones can enhance the cellular and humoral immuneresponses of mice in a concentration dependent manner. |
