Populus simonii Carr in6sites in Baoji and Yulin in Shannxi province were sampledfor endophytic fungi isolation by culturing sterilized leaves collected in Spring and Autumnrespectively, followed by investigation of endophytic fungi diversity and ecologicaldistribution. Results are as following:1. Strains of1160endophytic fungi were isolated from600leaf segments. Combinatedmorphological characteristics and molecular phylogenic technology, thirty five genues,twelvespecies were identified. Strains of1029endophytic fungi, taken on88.79%of total strains, aresubordinated to anamorphic ascomycetes, and they belong to three orders、five families、twenty-two genera, respectively; Strains of126belong to teleomorphic ascomycetes, taken on10.86%of total strains, and they belong to three orders、eight families、ten genera,respectively; Two strains are Zygomycota, belonged to1orders、2families、2genera,respectively; Two strains are Basidiomycetes, belonged to1orders、1families、1genera.Alternaria、Ulocladium and Phoma are dominant communities in Populus simonii, with RF23.10%,22.50%,14.22%, respectiely.2. Endophytic fungi in Populus simonii were rich in different loci. The amount, IR, CF inYulin is highter than that in Baoji, but index of Shaanon diversity is reversed. Index ofShaanon of P.simonii in these6sites ranged from1.5461to2.6202, and ranked by Fengnan,Taochuan, Yingge, Qinyun, Hongshan, Wuding Riverside as descent size. Coffient similarity(Cs) between Baoji and Yulin is0.4445. Cs within3loci of Baoji ranked from0.5000to0.5455, and0.6667to0.6897within3loci of Yulin. The highest Cs of0.6897among tested6loci occurred between Qingyun and Wuding Riverside, while, the lowest Cs of0.3889happened between Hongshan and Yingge. The similarity coffcient between Spring andSutumn in YingGe was highest (H’=0.5833), lowest between Spring and Autumn in QingYun(H’=0.4000).The Composition of endophytic fungi community was different between Spring andAutumn.The amount of isolates, colonization frenqucy (CF) and isolation rates (IR) inAutumn were higher than that in Spring, but the Shannon-Wiener index in Spring is higherthan that in Autumn. In Spring, Shannon-Wiener index of endophytic fungi communities in Hongshan is2.4875, the highest among the6sites, and that in Wuding riverside is the lowest,1.6918. In Autumn, Shannon-Wiener index of1.8988in Taochuang is the highest one, but thelowest one,1.0057, occurred in Fennan.3. The Componets of Endophytic Fungi community from Populus simonii showed closerelationship with geographical terrain、 climatic condition and ecological characteristi-cs.There was low diversity in Yulin, an arid region, but higher in Baoji, with theShannon-Wiener index in Baoji of3.4083compared to2.0625, which can be ascribe topercipitation and vegitation under the canopy of poplar stands.4. Molecular systematic of ITS-nrDNA is implemented here to help identify crypticspecies, and Cladosporium cladosporioides、 Cladosporium oxysporum、 Cladosporiumbruhnei、Cladosporium sphaerospermum、Cytospora chrysosperma、Valsa nivea、Eutypellascoparia、Thielavia subthermophila、Thielavia microspora、Glomerella cingulata、Phomaglomerata、Peyronellaea glomerata are figured out finally.
