Poplars are the wide spread species as well as among the fast growing trees on the earth. Because of our country large-scale territory, belongs to kinds of the climate and soil styles, so the poplar exceedingly rich. The researches on Poplar will have the significance of environment and economic value. In this paper, the extraction technology of the phenols from leaves of Populus Canadensis Moench., and the method for determination of IAA and GA3 in the male floral buds of Populus tomentosa Carr.were studied.Firstly, the extracting and deteminded methods t of total phenols from leaves of Populus canadensis Moench. were introduced. The single factor test and the orthogonal experimental design were carried out to optimize the extraction technology of total phenols from leaves of Populus canadensis Moench..The result indicated that the ethanol concentration, the ratio of solvent to material and the extraction temperature had a remarkable influence on the extraction of total phenols, while the extraction time had hardly influence on it. Extracting samples twice using ethanol as extraction solvent, result in the optimal conditions of extraction 70% ethanol and 40:1 the ratio of solvent to material, temperature of 55℃and extracting 10min. Using the above determined method,1.43% of total phenols was extracted. The optimized technology had good reproducibility and high precision.Secondly, effects of preservative on pork by the extractions from leaves of Populus canadensis Moench.were introduced. The single factor test is adopted to measure the changes o f smells, color and pH value with distilled water as controls.The results indicated that the extractions from leaves of Populus canadensis Moench.have certainly freshness function.Thirdly,the method for determination of IAA and GA3 in the male floral buds of Populus tomentosa Carr. were developed by high performance capillary electrophoresis. The samples were extracted with methanol for determination. The optimum determination conditions were investigated experimentally and the results were as follows:the running buffer of 10 m mol/L borate (pH=8.0), injection time with pressure of 5s, temperature of 25℃,applied voltage of 22 kV, detection wavelength of 200nm. Under the optimum conditions, a good linearity between peak area and concentration of IAA and GA3 were found. By above extraction technology, the IAA and GA3 content were 1.24μg/g andl.95μg/g. the analytes were completely separated in 10min.The analytical demonstrated the method is simple, fast and accurate.
