In this paper, in order to Yushu of Lamiophlomis rotata genetic diversity level,genetic structure and genetic differentiation, the geneticdiversity with Lamiophlomisrotata from10populations of Yushu area was analyzed by inter simple sequencerepeat (ISSR) molecular marker method. The meaning of the research is to providecertain theoretical basis for the development and protection of Lamiophlomis rotataresources. The main results of this thesis are as follows:The optimal ISSR-PCR reaction system for Lamiophlomis rotata was established.It is25μL reaction volume containing1.0U Taq DNA polymerase,1.5mmol/L Mg2+,75ng templates DNA,0.05mmol/L dNTPs and0.75μmol/L primer. The optimalamplification procedures were per-denaturing for5min at95℃, followed by40cycles of denaturing for40s at95℃, annealing for40s, extension for90s at72℃.Then extension step of7min at72℃was performed.In this study,18ISSR primers were screened for the PCR amplification ofLamiophlomis rotata,188bands were resulted, of which187were polymorphic bands,and the percentage of polymorphic loci was99.47%. POPGENE analysis indicatedthe genetic diversity of Lamiophlomis rotata at the species level (PPL=99.47%, Ne=1.4777, Nei=0.2904, I=0.4494).By processing the data, draw relevant data for genetic structure: Lamiophlomisrotata from10populationds of Yushu(Ht=0.2904, Hs=0.2180, Gst=0.2493, Nm=1.5055). Nei’s genetic distance (D) of Lamiophlomis rotata populations is between0.0076and0.2061, the average of Nei’s genetic distance (D) is0.1052. The show ofUPGMA cluster analysis of Nei’s genetic distance by MEGA5is that ZY population,HB population, XQ population and CY population is a group, but HT population andSP population is another group, BXpopulation, BZ population and CF population is agroup, at last, only CD population is a group.Comprehensive analysis, Yushu area Lamiophlomis rotata population geneticdiversity showed high diversity in the overall and average, the relatively lowgeneticdiversity and individual population revealed, is likely to be human activitiesandnatural disasters, need specific reasonable measures to protect.
