In China, the promotion and application of boar semen cryopreservation technology hasbeen slow, mainly due to the viability and the rate of reproductive of the artificialinsemination of frozen sperm is not ideal. In order to further improve the semen frozen-thawed quality and resistance to low temperature injury blow, in this experiment, we usedthe pachymaran, APS and Ginkgo flavones as freeze protection additive, using PI,rhodamine123,HOST and FITC-PNA to detect the sperm motility, mitochondrial activity,acrosome and membrane integrity, and evaluating the protective effects of semen quality.Meanwhile, we detectd the effect of SOD, LDH and GOT enzyme activity of pachymaran,APS and Ginkgo flavones on sperm freezing and thawing, the following results wereobtained.1. During freezing sperm, the traditional frozen on the basis of dilution TCG were addedPachymaran0.06mg/mL the APS0.04mg/mL and0.08mg/mL of Ginkgo flavonoids canimprove the survival rate of boar sperm after freezing, the rate of plasma membrane integrity,acrosome integrity rate and mitochondrial activity (P <0.05). Three kinds of cryoprotectantscompare,0.06mg/mL of poria cocos protection effect is good, compared with control group,live sperm rate increased by50.15%, acrosome intact rate increased by66.41%, increased by21.27%, plasma membrane integrity mitochondrial activity increased by10.93%.2. Selection of different concentrations of pachymaran, APS and Ginkgo flavonoids withdifferent concentrations of glycerol crossover design of two factor variables, which after0.06mg/mL Pachymaran and2.4%glycerol with sperm motility, mitochondrial activity,membrane integrity and acrosome integrity rate were best. compared with the control groupsignificant difference (P<0.05), compared with when glycerin content for3%of theexperimental group difference was not significant (P>0.05), the results showed that the sugarwith glycerol can effectively improve the quality of the boar sperm after frozen, Pachymaranof right amount add can effectively reduce the amount of adding glycerol, but they can notcompletely replace the glycerin used as antifreeze protection.3. After frozen-thawed, the activity of superoxide dismutase of boar sperm (SOD)decreased, while lactate dehydrogenase (LDH) and aspartate transaminase (GOT) in thefreezing process is gradually elevated; after adding cryoprotectants, the activity of SODincreased, which0.06mg/mL pachymaran protective effect of SOD on the best balance of the activity of105.26u/mL, frozen activity81.04u/mL. After adding refrigerant, GOT and LDHactivity increased more slowly than the control group.
