To explore the different distribution in different tissues and cells of neuroglobin betweenadult yak and cattle, Eight health adult yaks were used in this study.NGB expression indifferent areas (cerebral cortex,cerebellar cortex, hippocampal, striatum, medulla oblongata,olfactory bulb of nervous system and peripheral nervous system;,pituitary,adrenal, pineal gland,thyroid of endocrine system; testis, ovary and fallopian tube of reproductive system; abomasumand duodenal glands jejunum, ileum, cecum, colon, pancreatic acinar cells and ductal epithelialcells of alimentary system and cardiac muscle, skeletal muscle, liver, spleen, lungs, kidneys andother organs and tissues) of the adult yak and cattle brain was localized using theimmunohistochemical SP technique and NGB expression in different areas (cerebral cortex,cerebellar cortex, medulla oblongata, striatum, olfactory bulb, adrenal gland, abomasum,duodenum tissue) of the adult yak and cattle brain was localized using the real-time fluorescentquantitative PCR technology.The research results show that:1. The immunohistoche mical results1.1Distribution of neuroglobin in nervous system of adult yak and cattleIn cerebral cortex of yak, all the Ⅰ~Ⅵ layer of cerebral cortex have positive cellsdistributed, and the strength of cerebral cortex was significantly higher than the region of thecerebellar cortex, hippocampus and striatum. NGB positive cells distributed in the medulla werealso observed. In cerebral cortex of cattle, the distribution and localization of NGB positive cellswas similar with yak, and the strength of cerebral cortex was significantly higher than the regionof the cerebellar cortex, hippocampus and striatum. NGB positive strength in the cattle’s brainwas overall weaker than yak.In cerebellar cortex of yak, NGB positive cells was stronger in purkinje s cell layers, andwas weaker in molecular and granular layers. In cerebellar cortex of cattle, the distribution andlocalization of NGB positive cells was similar with yak, but the intensity of reaction was overallweaker than yak.In different parts of the hippocampal in yak, NGB positive cells were mostly pyramidalcells, NGB positive reaction site was located in the nerve process and weaker. In different partsof the hippocampal in cattle, the distribution and localization of NGB positive cells was similarwith yak, but the intensity of reaction was overall weaker than yak.In the striatum of yak, NGB positive cells were widely distributed in the region of thecaudate nucleus, and positive reaction was stronger than the region of hippocampus, medullaoblongata, olfactory bulb, weaker than the region of cerebral cortex, cerebellar cortex. In thestriatum of cattle, the distribution and localization of NGB positive cells was similar with yak, the intensity of reaction of NGB positive cells in the caudate nucleus was higher thanhippocampus, medulla oblongata, olfactory bulb, and was overall weaker than yak.In the medulla oblongata of yak, NGB positive cells were mainly distributed in the graymatter, NGB positive cells in the white matter was also scattered. In the medulla oblongata ofcattle, the distribution and localization of NGB positive cells was similar with yak, but theintensity of reaction was overall weaker than yak.In the mitral cell layer of yak olfactory bulb, the staining was significant and cells were large,the staining intensity of NGB positive cells was weaker than medulla oblongata and higher thanhippocampus. In the mitral cell layer of cattle olfactory bulb, the distribution and localization ofNGB positive cells was similar with yak, the staining intensity was higher than hippocampus andweaker than medulla oblongata, and was overall weaker than yak.In the peripheral nervous system of yak was similar with cattle. NGB positive cells weremainly distributed in the peripheral nerve plexus and ganglion with mostly scattered in some ofthe nerve cells in the peripheral nervous system, and the quantity was fewer. In the peripheralnervous system of cattle, the distribution and localization of NGB positive cells was similar withyak, but the intensity of reaction was overall weaker than yak.1.2Distribution of neuroglobin in endocrine systemof adult yak and cattleIn the yak adrenal, NGB was distributed in the cortex and medulla. In the cattle adrenal, thedistribution and localization of NGB positive cells was similar with yak, and there was nosignificant difference in the intensity of reaction between yak and cattle.In the yak pituitary, NGB positive cells were mainly eosinophils, basophils andchromophobe. In cattle pituitary, the distribution and localization of NGB positive cells wassimilar with yak, and there was no significant difference in the intensity of reaction between yakand cattle.The adult yak and cattle pineal gland, thyroid and other organs and tissues were observedusing immunohistochemical methods. NGB positive cells were not found.1.3Distribution of neuroglobin in reproductive system of adult yak and cattleIn yak testis, the coloring of NGB positive cells in spermatogonia and primaryspermatocytes was deepest, the coloring in other spermatocytes and part of the interstitial cellswas slightly lighter. In cattle testis, the distribution and localization of NGB positive cells wassimilar with yak, and there was no significant difference in the intensity of reaction between yakand cattle.Ovary and fallopian tube in the female reproductive system of adult yak and cattle wereobserved using immunohistochemical methods. NGB positive cells were not found.1.4Distribution of neuroglobin in alimentary system of adult yak and cattle In the abomasum of yak, NGB positive cells were in the fundic glands cells of abomasums.In the abomasum of cattle, the distribution and localization of NGB positive cells was similarwith yak, and there was no significant difference in the intensity of reaction between yak andcattle.In the duodenum of yak, NGB positive cells were in the duodenal gland cells. In theabomasum of cattle, the distribution and localization of NGB positive cells was similar with yak,and there was no significant difference in the intensity of reaction between yak and cattle.Jejunum, ileum, cecum, colon, pancreatic of adult yak and cattle were observed usingimmunohistochemical methods. NGB positive cells were not found.1.5Distribution of neuroglobin in other issues and cells of adult yak and cattle.The adult yak and cattle cardiac muscle, skeletal muscle, liver, spleen, lungs, kidneys andother organs and tissues were observed by using immunohistochemical methods. NGB positivecells were not found.2. The fluorescence quantitative resultsIn diifferent tissues of yak, The adult yak cerebral cortex, cerebellar cortex, medullaoblongata, striatum, olfactory bulb, adrenal gland, abomasum, duodenum tissue of NGB genes2-△△Ctvalues were262.69,137.00,3.43,7.65,2.14,7.82,16.26and30.23. The expresion ofNGB gene in the cerebral cortex, cerebellar cortex, medulla oblongata, striatum, olfactory bulb,adrenal gland, abomasum, duodenum tissue were262.69times,137.00times,3.43times,7.65times,2.14times,7.82times,16.26times and30.23times than the expression of hippocampus.Results are consistent with the results of immunohistochemistry. The expression of NGB gene inthe cerebral cortex, cerebellar cortex was significantly higher than that of other organizations,there was the lower expression in the duodenum and abomasums, following by the expression ofthe adrenal gland, striatum, there was the lowest expression in the medulla oblongata, olfactorybulb, hippocampus. In different tissues of cattle, The adult yak cerebral cortex, cerebellar cortex,medulla oblongata, striatum, olfactory bulb, adrenal gland, abomasum, duodenum tissue of NGBgenes2-△△Ctvalues were165.53,136.29,1.53,1.98,2.19,4.18,12.91and20.29. Theexpression of NGB gene in the cerebral cortex, cerebellar cortex, medulla oblongata, striatum,olfactory bulb, adrenal gland, abomasum, duodenum tissue were165.53times136.29times,1.53times,1.98times,2.19times,4.18times,12.91times and20.29times than the expression ofhippocampus. The expression of NGB gene in the cerebral cortex, cerebellar cortex was thehighest and significantly higher than in other tissues, there was the lower expression in theduodenum, and abomasums, there was the lowest expression in the adrenal gland, striatum,medulla oblongata, olfactory bulb, hippocampus.
