Phosphorus is the major limiting factor for plant growth and yield. Saline-alkaline soils causes the low fertilizer efficiency of phosphate, and the poor application effect of the inoculant of phosphate-solubilizing bacterias(PSB) in Tarim Basin, Xinjiang. Hence it is very important for the application of PSB in specific region that screening and identification of high efficiency PSB as inoculant to manufacture bio-fertilizer.In this study, the grape rhizosphere soil samples was gathered in the main grape planting region(Shanshan, Aksu, Hetian Yanqi, and Heshuo) of Tarim Basin in Xinjiang with various varieties and tree-ages. The objective of the study was to explore the distribution of PSB, screen and identify of high efficiency PSB by physiological, biochemical and 16 S rDNA method, the main conclusions are as follows:1.There are a large number of PSB in Tarim Basin, including 263 inorganic PSB strains and 226 organic PSB. The distributions of PSB in different areas, various varieties and different tree-ages soils present a certain degree of difference. The significant positive correlation was found between the amount of PSB and alkali-hydrolyzable nitrogen.2.The performance of solubilizing inorganic phosphorate was range from 94.64 μg·mL-1 to 1330.66 μg·mL-1, and 42 strains had high phosphate solubilizing capacity(PSA) greater than 700 μg·mL-1, the PSA of organic PSB was from 1.64 μg·mL-1 to 52.02 μg·mL-1, with 22 strains higher than 30 μg·mL-1.3.Combined with cell morphology observation, physiological and biochemical identification and 16 S rDNA sequence analysis, inorganic PSB belonged to 5 genera and 10 species in which 18 strains belonged to Serratia sp., including Serratia liquefaciens, Serratia plymuthica, Serratia proteamaculans, 14 strains belong to Pseudomonas brassicacearum, another 5 strains belonged to Bacillus subtilis, Klebsiella oxytoca, Enterobacter aerogenes, Enterobacter cloacae, Enterobacter asburiae, respectively. And orgnanic PSB belonged to 7 genera and 7 species, 5 strains belong to P. brassicacearum, 2 strains belong to Bacillus amyloliquefaciens, 2 strains belong to Stenotrophomonas maltophilia, 2 strains belong to Serratia sp., another 3 strains belong to E. cloacae, Agrobacterium sp. and Advenella kashmirensi respectively.
