| Plant regulating many function genes of drought stress response and lead to many morphological structure and physiological and biochemical changes to adapt to drought stress environment, transcription factor plays an extremely important role in regulating the expression of the function genes. It has great application value and significance to supplement and improve the wheat drought resistance identification system by exploring transcription factor genes closely related to drought stress response as molecular indexes of wheat drought resistance identification.This study using real-time quantitative PCR technology, tested the expression of four drought stress response relative transcription factor genes Ta ERF1, Ta MYB30, Ta NAC69 and Wabi5 in four different wheat varieties, and measured the change of four commonly used physiological and biochemical indexes of drought resistance identification in each wheat varieties, further analyzed the correlation between the expression of the genes and the change of the physiological and biochemical indexes, discussing feasibility of taking the expression of Ta ERF1, Ta MYB30, TaNAC69 and Wabi5 as the index of wheat drought resistance identification. Our study obtains these main conclusions:(1) After PEG stress, the expression level of Ta ERF1, Ta MYB30, Ta NAC69 and Wabi5 in different wheat varieties are all raise, and the increase of the expression level in the drought resistance varieties are greater. The expression time and patterns of TaERF1, Ta MYB30, TaNAC69 and Wabi5 are differences under drought stress. These results indicated that transcription factors Ta ERF1, TaMYB30, Ta NAC69 and Wabi5 associated with the drought stress response of wheat, they regulate target genes as positive regulatory factors after wheat under drought stress.(2) After PEG stress, the regulation ability of drought related gene’s expression and the physiological and biochemical indexes in drought resistance wheat varieties are stronger, and there are correlation between expression of the genes and the change of physiological and biochemical indexes. After PEG stress, the change of soluble protein and soluble sugar in drought resistance wheat were bigger and the change of MDA and RWC were smaller, this is most obvious in drought resistance wheat varieties Pubing9946, so Pubing9946 must be a good material for wheat drought resistance breeding research.(3) The expression of Ta ERF1, Ta MYB30, Ta NAC69 and Wabi5 is induced by drought stress signals, through correlation analysis, the expression of them exist significant correlation with the change of common drought resistance identification indexes soluble protein, soluble sugar, MDA and RWC, showed that the expression of TaERF1, Ta MYB30, TaNAC69 and Wabi5 can be used as a reference indexes of wheat drought resistance identification. |
