| Wheat is one of the main grain crops in our country,and its yield is directly related to grain security,but often affected by drought.So it is important to explore drought resistant genes and breed new drought resistant varieties by molecular breeding.And studies have shown that transcription factor genes are important candidate genes for drought resistance.In the previous studies on gene resistance,we screened two nuclear transcription factor gene of NF-Y family which are TaNF-Y1 and TaNF-Y2 from the drought transcription spectrum of wheat.And preliminary functional tests in Arabidopsis showed that these two genes were able to improve drought resistance in transgenic arabidopsis.In order to further verify their function in wheat,we obtained transgenic wheat T5 lines with TaNF-Y1 transgenic wheat S4185 and TaNF-Y2 transgenic wheat J22 respectively.On this basis,we identify the drought resistance of two kinds of transgenic wheat with the physiological drought related biochemical indexes and yield as the basis at the stress field in the indoor drought and water control condition,which analyzed the drought resistance function of TaNF-Y1 and TaNF-Y2 genes in wheat.The main results are as follows:Three TaNF-Y1 transgenic lines were significantly higher than the recipient cultivar "stone 4185" in germination vigor,germination rate,seedling biomass and total root length of germinating seeds under the same PEG simulated drought stress condition,and dehydration rate of detached leaves and relative electrical conductivity of seedlings were just the opposite,which was good for water conservation.More importantly,the yield of three transgenic lines was significantly higher than that of the recipient cultivar "stone 4185" under the control of field water and normal irrigation,And under water control condition,the output increase is greater.These results indicated that overexpression of TaNF-Y1 gene in recipient cultivar "stone 4185" was beneficial to drought tolerance in wheat.Comparing with the recipient cultivar "Jimai 22",the yield of the three TaNF-Y2 transgenic lines was significantly higher under field water control and normal irrigation,and the increase of yield was mainly due to the increase of 1000 grain weight.Grain filling stage is the key period for the formation of 1000 grain weight,in this period,MDA content and relative conductivity of leaves of three TaNF-Y2 transgenic lines was significantly lower than the recipient cultivar "Jimai 22" under indoor water treatments,while soluble protein content was significantly higher;The results of indoor water control at booting stage are consistent with that at filling stage.The above results showed that overexpression of TaNF-Y2 gene in the recipient cultivar "Jimai 22" was beneficial to enhance the ability of wheat to resist drought stress. |
