Artificial insemination is widely used in the modern pig industry to improve the production efficiency, and the boar semen dilution powder is a key factor of the quality of semen preservation because artificial insemination usually choose boar semen stored at room temperature. Currently, the boar semem normal temperature preservation dilution powder products is mainly depend on imports, the price is higher, the quality is different from each other, so that develop a diluted powder products with independent intellectual property is of great significance to promote the development of the pig industry in China. In order to improve the formula of boar semen diluted powder preservation at room temperature and improve the effect of the diluted powder, this research improved the dilution based powder formula by adding 1.6 g/L, 1.8 g/L, 2.0 g/L, 2.2 g/L β-cyclodextrin in the basic extender of previous studies. After adding appropriate amount of β-cyclodextrin, we studied the effects of vitamin C and vitamin E on normal temperature preservation of porcine semen. Finally,according to the improved dilution powder component, small batch pilot products were used in modern hoggery, simultaneously carried on the contrast experiment with imported diluent.The results of this research are as follows:1. Different concentrations(1.6, 1.8, 2.0 and 2.2 g/L) of β-cyclodextrin were added in basic dilution powder of boar semen, respectively, in each group of withen the increasing of the concentrations of β-cyclodextrin, acrosome integrity, plasma membrane integrity and motility were improved. Motility and acrosome integrity of the group added 2.0 g/L was significantly higher than other group(P < 0.05), plasma membrane integrity of group added1.8 g/L and 2.0 g/L showed no significant difference(P < 0.05); The optimum concentrations of β-cyclodextrin in basic extender was 2.0 g/L, and motility was 58.00%, plasma membrane integrity was 57.00%, and acrosome integrity was 68.57% after 5 days of preservation.2. The basic dilution powder with 2.0 g/L β-cyclodextrin, the group added 10 mg/L vitamin C and other significant difference to other group(P < 0.05). After 5 days preservation,sperm motility was 62.00%, plasma membrane integrity was 54.80%, acrosome integrity was67.00%, H2O2 concentrations was 85.10 mmol/mL, T-AOC concentrations was 3.83 IU/ml,and MDA content was 1.30 nmol/ml. The group added 25 mg/L vitamin E was significantdifference to other groups(P < 0.05). After 5 days preservation, sperm motility was 62.00%,plasma membrane integrity was 52.50%, acrosome integrity was 67.50%, H2O2 content was93.77 mmol/mL, T-AOC activity was 3.90 IU/m L, and MDA content was 1.40 nmol/m L.3. The diluent of “Pregnancy Benefits”(the best formula of Experiment 1) and the commercial diluent was used to do the breeding test respectively in two varieties of Landrace and Large White boars respectively. Since with thinner in Landrace sow mating experiment,litter number born alive, and abnormal rate, weak rate and alive with the thinner, the results showed that the experimental group were significant difference(P < 0.05). In the group of Landrace with the diluent of “Pregnancy Benefits”, the conception rate was 94%, birth litter weight was 15.18 kg, litter number born alive was 10.78, weak abnormal rate was 1.32%,Live birth rate was 93.69%, and newborn weight 1.42 kg. Since with thinner sow mating experiment in the litter number born alive, the abnormal rate of weak China, alive rate and initial weight and commodity thinner, experimental group were significant difference(P <0.05). In the group of Large White boar, the diluent of “Pregnancy Benefits”, the conception rate was 95.33%, birth litter weight were 15.73 kg, litter number born alive was 10.62, weak abnormal was 2.78%, live birth was 92.51%, and newborn weight 1.49 kg. The effect of diluent of “Pregnancy Benefits” on boar semen had no significant difference compared with the commercial diluent, and the diluent of “Pregnancy Benefits” can be applied in production.
