The Genetic Linkage Map Construct Ion Of Apple Based On SLAF Marker

Apple is one of the most important deciduous fruit trees in the world. At now, the research on genetics and molecular biology of apple germplasm resource have been the hotspot both in china and oversea and the use of advanced molecular marker assisted breeding technology has become an important trend in apple breeding, A fine genetic map is the precondition for apple traits directional improvement.‘Fuji’ and ‘Qinguan’ apple both have their own special excellent characters respectively, and their have an important position in the apple cultivation and breeding of China. Their have been used as important parent material in the cross breeding. In this study, with the F1 population of 158 individuals crossed from ‘Qinguan’ and ‘Fuji’ apple, the SLAF tag between the parents were developed, and the high density genetic linkage map of Qinguan and Fuji were constructed using SLAF markers. The main results were as following:1. In total, 321,068 SLAF labels were developed, among them, 125,497 were polymorphism SLAF tags, and 58,159 SLAF tags can be used in genetic map construction, counted to 18.11% of the total tags. At last, 5,485 SLAF markers were placed on the genetic map.2. A high detisty and netural genetic map of ‘Qinguan’ and ‘Fuji’ was constructed, containing 17 linkage groups and 5,485 SLAF markers, spaning 3,136.63 cM with an average distance of 0.57 cM between consecutive markers. The female Qinguan apple map having 3,132 SLAF markers, spaning 2,909.29 cM with an average distance of 0.93 cM between markers. The male Fuji apple locaed 3,255 SLAF markers, spaning 3,299.87 cM with an average distance of 0.99 cM between consecutive markers.
