Fokienia hodginsii (Dunn) Henry et Thomas is a single plant of the genus Fokienia hodginsii. It’s an ancient relic plants and endangered species in our country and it has the landscape application value. This study analysis genetic diversity of Fokienia hodginsii (Dunn) Henry et Thomas of 35 provenance samples with 5 province by the ISSR molecular markers and improved variety selection by using the analytic hierarchy process (ahp).The main research results get following conclusion:1、The 20μl ISSR-PCR reaction system that suitable for Fokienia hodginsii (Dunn) Henry et Thomas contains 2μl IOxPCR buffer,1.8 mmol·L-1 Mg2+,0.25 mmol·L-1 dNTPs,0.25U Taq DNA polymerase, 0.7μmol·L-1 of primer and 50ng template DNA. PCR program: predenaturing 5min at 94 ℃, then run 30s at 94℃,40s at 48.6℃ and 2min at 72℃ for denaturing, annealing and extension for 40 cycles and run 7min at 72℃ for extension.2、The ISSR amplifications for 45 samples of Fokienia hodginsii (Dunn) Henry et Thomas by 17 primers screened from 100 ISSR primers detect 237 loci in total, among which 217 loci are polymorphic, accounting for 91.56% of the lotal loci. Each primer amplification loci are between 9 to 19 and with an average of 13.94. Ploymorphism loci are between 8 to 18 and with an average of 12.76. Various primers to detect the DNA fragment size are between 200bp to 4500bp.3、45 samples of Fokienia hodginsii (Dunn) Henry et Thomas was constructed based on ISSR DNA fingerprint analysis by using the primers UBC854, UBC849, UBC857, UBC860 and UBC840.4、By using software Popgene 32 to analysis 45 samples of Fokienia hodginsii (Dunn) Henry et Thomas as results:The number of observed alleles is 1.9156, the efficient alleles is 1.4348, the index of genetic diversity is 0.2610, the Shannon’s information index is 0.4016. the total gene diversity is 0.2610. The genetic similarity is between 0.5949 to 0.8481 and with an average of 0.7492, the genetic distance is between 0.1499 to 0.5193 and with an average of 0.3078.5、35 provenances of Fokienia hodginsii (Dunn) Henry et Thomas’ percentage of polymorphic loci is between 26.16% to 54.43% , with an average of 37.67%. The number of observed alleles is 1.8312, the efficient alleles is 1.4076, the index of genetic diversity is 0.2444, the Shannon’s information index is 0.3753, the total gene diversity is 0.2444. The genetic similarity is between 0.6203 to 0.8608 and with an average of 0.7540, the genetic distance is between 0.1499 to 0.4776, with an average of 0.2914. The genetic relationship between provenances tree farm of QuanZhouFengTian and tree farm of QuanZhouZhuYuan is the most recent and the genetic relationship between provenances tree farm of SanMingYouXi and natural reserve of ShaoGuanLuoKen is the farthest.6、7 provenances of Fokienia hodginsii (Dunn) Henry et Thomas in FuJian Province detect 187 loci which are polymorphic.The average polymorphism rate is 78.90%. The polymorphism loci is between 36 to 150 and with an average of 88. The percentage of polymorphic loci is between 15.19%to 63.29%and with an average of 37.07%. The number of observed alleles is 1.7890, the efficient alleles is 1.3517, the index of genetic diversity is 0.2150, the Shannon’s information index is 0.3337, the total gene diversity is 0.2072, the gene diversity within populations is 0.1320, the coefficient of gene differentiation is 0.3629 and the gene flow is 0.8780. The genetic similarity is between 0.8324 to 0.9523 and with an average of 0.9038, the genetic distance is between 0.0489 to 0.1834 and with an average of 0.1047.7、5 provenances of Fokienia hodginsii (Dunn) Henry et Thomas’polymorphism loci is between 59 to 187 and with an average of 111. The number of observed alleles is 1.9156, the efficient alleles is 1.3861, the index of genetic diversity is 0.2352, the Shannon’s information index is 0.3663, the total gene diversity is 0.2410, the gene diversity within populations is 0.1583, the coefficient of gene differentiation is 0.3432. The genetic similarity is between 0.8412 to 0.9436 and with an average of 0.8900, the genetic distance is between 0.0581 to 0.1729 and with an average of 0.1414.8、According to UPGMA clustering method,35 provenances of Fokienia hodginsii (Dunn) Henry et Thomas were divided into four categories. The first class is the provenances of No.32, the second class is the provenances of No.10, the third class is the provenances of No.2, No.5, No.7, No.9, No.11, No.16, No.22, No.33, No.34 and No.35, the last class is the remaining provenances.7 provenances of Fokienia hodginsii (Dunn) Henry et Thomas in FuJian Province were divided into three categories. The first class is the provenances of NanPin City, the second class is the provenances of ZhangZhou City and the third class is the provenances of QuanZhou City, SanMing City, FuZhou City, LongYan City and NingDe City.5 provenances of Fokienia hodginsii (Dunn) Henry et Thomas in 5 provinces were divided into three categories. The first class is the provenances of FuJian Province, GuangXi Province and HuNan Province, the second class is the provenances of GuangDong Province and the last class is the provenances of SiChuan Province.9、The analytic hierarchy process is adopted to establish an evaluation system of landscape type of Fokienia hodginsii (Dunn) Henry et Thomas and the evaluation factor and its weight is determined. According to this system,16 trees were selected in the experiment to evalution, the results show that the Ⅰ level include No.10 and No.11. The Ⅱ level include No.3, No.9, No.12 and No.14. The Ⅲ level includes remaining samples.
