Tomato yellow leaf curl disease became one of the most prevalent ang damaing diseases in tomato production in the world. At present, the disease has occurred in large scale in China’s major tomato cultivation areas for tomato production has brought serious harm or even destructive. Use in protected more than 60 mesh gauze physical isolation, and spraying pesticides from sowing, planting to harvest the whole process of the spread of TYLCV whitefly can control, so as to achieve the purpose of control TYLCV. But requires a lot of investment and a lot of pesticides, and open field tomato difficult physical isolation, the use of pesticides to control only difficult to effectively control TYLCV.Therefore, the anti-virus Variety tomato breeding has become one of the most important breeding objectives. So far, five resistance genes have been mapped, i.e. Ty-1, Ty-2, Ty-3, Ty-4 and Ty-5, derived from several wild tomato species. By combining field with molecular markers has bred some resistant varieties, mainly containing Ty-1 and Ty-3 gene. Ty-2 gene on China’s major tomato yellow leaf curl virus resistant to race, therefore, the Ty-2 resistance gene fine mapping and cloning will have extremely significance to functional analysis and breeding tomato varieties. In this study, on the basis of existing, The cultivated tomato line 12g-60, Lycopersicon Tomatoes contain the same location on a chromosome fragment susceptible introgression lines LA4006 and F2 hybrid population as the main research material fine mapping the Ty-2 gene, BAC library construction, gene prediction and other research, the main results are as follows: 1. Using the cultivated tomato line 12g-60, Lycopersicon Tomatoes contain the same location on a chromosome fragment susceptible hybrid introgression lines LA4006 hybrid and get the corresponding F2 populations.The use of poison whiteflies in the F2 populations for natural inoculation. At the same time,taking advantage of five molecular markers for genetic analysis of the 1320 plants population. The results showed that the anti-sense ratio is 3:1,in accordance with the identification results of single gene inheritance,disease resistance results and molecular marker results are consistent 2. The cultivated tomato line 12g-60 was selected for the construction of bacterial artificial chromosome(BAC) library. Four BAC clones were detected by PCR. The series number of them were F17F24, F18E2, F118A13, and F187N19. These four clones were sequenced full-length showed that there is an inversion between the common tomato and S.habrochaites chromosome fragment of the Ty-2 gene at the same position, Thus explains the serious restructuring suppression occurring in Ty-2 gene fine mapping studies. 3. The use of bioinformatics tools for four positive clones sequences predicted candidate genes.There are 21 genes present on the chromosome fragment purpose.These gene mainly code 2-phosphoglycerate kinase, chaperonin, cytochrome P450, ABC transporter G family member 28.There was only one gene Solyc11g069620.1 which contained the conserved domains of NB-ARC class and located in the recombinant area, were selected as Ty-2 candidate genes, Transgenic research is underway...
