The main way to overcome the contradiction between the quality and yield of rapeseed is heterosis,and self-incompatibility is one of the ways of heterosis utilization,and breeding self-incompatibility lines are the key. The experiment used molecular markers to identify the parental genotypes and breed auxiliarily self-incompatible BC1 groups and F2 groups.The following main results:1.The S-haplotype identification of 280 Brassica napus males were 155 restorer lines in 55.36% and 82 maintainer lines in 29.29%,others were heterozygous strains.Brassica mostly are restorer lines.2.According to the results of genotyping 57 F2 populations materials which were 3100 plants in total, 601 homozygous Bns-1300 genotype were bagged.Observed the phenotype showed that 499 plants were self-incompatibility and 102 plants were self-incompatibility.So self-incompatible plants accounted for 83.03%.It can be concluded that the detection results of molecular marker were same with phenotypic of field.3.The 82 self-incompatible F2 plants with good selection were carried out by the stripping and budding,and the F3 seeds were obtained.4. According to the results of genotyping 42 BC1 populations materials which were 810 plants in total, 346 homozygous Bns-1300 genotype were bagged.Observed the phenotype and selected 35 plants with good traits of heterosis to be remained, few which could not self-seed would be stripped bud selfing, and BC1F2 seeds were obtained.5. Optimizing molecular marker technology and developing dual-PCR technology to identify rapeseed three lines, the results showed at some temperature these primer combinations could not run out of some dual band, some could not be specific. Among all cases with DNA bands, optimum temperature with dual band and specific could not be found.These results suggest that the molecular marker system for breeding of selfincompatibility lines is effective and feasible.The selected self incompatible materials are helpful for the further breeding of the self-incompatibility lines and the self incompatible hybrids.
