Self-incompatibility is one of the most important pollination systems for hybrid seed production in B. napus,And it’s an urgent thing to improve the existed SI lines.In this study, S-1300, a B. napus self-incompatible line and103accessions collected from China and foreign countries were used as parent materials, in which the aim was to analyze the genetic control of self-incompatibility in rapeseed. And the molecular markers which are closely linkage with self-incompatibility gene are used to assist select B. napus self-incompatible lines. The major results were as follows:1. one F2population (510plants) generated from " S-1300×9-9600" and one BC1population (40) plants generated from "(S-1300×9-3600)×S-1300" were investigated for self-incompatibility/self-compatibility. In F2population, there were369SC plants and141plants;In BC1population, there were22SC plants and18SI plants, as it was expected according to Mendelian segregation3:1and1:1. The results suggested that self-incompatibility in S-1300was under the single locus control.2. In F2(21plants) population generated from "S-1300×9-6266(Restorer)"the genotype S47S47、S60S47、S60S60were6plants、7plants,8plants, the revelant phehotype SC、SC、SI were6plants、7plants、8plants; In F2(21plants) population generated from "S-1300×9-6265(Maintainer)", the genotype S60S60、S60S29、S29S29were6plants、9plants、3plants (3samples were not be tested), the revelant phehotype SI、SI、SC were6plants、8plants、7plants; In homozygous S-1300line, the tested genotype were all S60S60, The phehotype were all SI. These indicated the molecular makers that were designed for this study are effective in MAS3. In this study, S-1300(Brissica napus L.) as female parent and other accessions as the male parents to get the hybrid F1, then by backcross with the male parent to get the BC1, from which select the heterozygous type by molecular markers, followed by continuous selfing and molecular markers assisted selection, We selected the pure self-incompatible individual propagated by selfing, artificial pollination till BC1F3, combined with phenotypes appraisal, we can obtain the improved SI lines.4. Also, S-1300(Brissica napus L.) as female parent, but different accessions as the male parents to get the hybrid F1, follow by continuous selfing and molecular markers assisted selection in F2segregation population, the pure self-incompatible individuals were selected and propagated by selling, artificial pollination till F4, combined with phenotypes appraisal, we can also get the improved SI lines.
