AbstractGene silencing is a kind of natural antiviral mechanism,The product of the gene SGS3 is a core protein of plant PTGS process, At SGS3 and SISGS3 have been proven to be of natural antiviral function. This research is based on a Bn SGS3 gene found in Brassica napus : Using the Southern blot hybrid defined copy numbers of the Bn SGS3 in different rapeseed; Bn SGS3 gene over expression vector and interference expression vector were transformed into arabidopsis thaliana by way of floral-dip mediated by agrobacterinm tumefaciens and carries on the virus resistance identification;Through inoculate Botrytis cinerea and Sclerotinia disease With rape plant interference expression of the Bn SGS3, clear whether the interference of the Bn SGS3 gene expression affected the broad-spectrum disease resistance in rapeseed, to explore the Bn SGS3 expression changes on the influence of the rape plant growth and development.The main results are as following:1、Bn SGS3 copy number is 1 in the non-GMO of zhongshuang six and suyou one plant;Bn SGS3 over expression and interfere expression of transgenic rapeseed all show different copy number per plant, but the highest do not exceed 3, the lowes is one.2、In different varieties of arabidopsis vaccine virus experiment, Over express Bn SGS3 transgenic arabidopsis thaliana only for CMV showed resistance;Interference expression Bn SGS3 transgenic arabidopsis did not show differ disease resistance between the wild type arabidopsis.3、Arabidopsis thaliana plants and oilseed rape plants transformed in Bn SGS3 compared with normal plants have great differences in phenotype,These transgenic plants alter on phenotype not only change virus resistance,changing its resistance to other diseases;Fungal inoculation on Bn SGS3 interference expression in rapeseed plants experiment, transgenic plants relative to the control group showing symptoms of the disease is aggravating.
