Study Of Breeding Population Performance Measurement And Microsatellite Marker Of Genetic Structure On Shenxian Pigs

As once local varieties of hebei province, shenxian pigs was deeply loved by local people.but adversely affected by imported lean hogs.In 2004,When investigating national indigenous pig breeds,because of the amount small,so that unable included in the local chronicles.In 2009,start to collect shenxian pigs from the folk of Xin JI Shi Zheng Nong Mu Ye Co.,ltd.After five years of collecting,excavating, purification and rejuvenation,shenxian pigs herd has begun to take shape.For understand and master performance and genetic structure of shenxian pigs herd..So we beginning this study.This study is divided into two parts: part one is the population performance measurement,part two is the Microsatellite Marker of Genetic Structure on Shenxian pigs.Test one :This experiment studied the Shenxian pigs herd by collect and form in hebei province,and finally settled down Xin JI Shi Zheng Nong Mu Ye Co.,ltd.Record the Reproductive performance indicators,like litter size, birth weight, weaned weight, survival rate, etc.Record the growth performance indicators,like body weight and body size.Record the indexes of slaughter trai,like carcass lean meat percentage, crude protein, crude fat, PH, drip loss and the meat color, etc.Record the Meat quality indexes such as the thickness and density of muscle fiber. Size appearance, the growth, meat quality characteristics, Slaughter performance of shenxian pigs were determined in the study.At present,boar population 24,sows 147 of shenxian pigs.First-born litter size is 9.23,The average of piglet birth weight is 0.94 kg,weaning weight is 5.57 kg,eye muscle area is 21.20cm2,lean rate is 35.08%,muscle colour points is 3.5,Muscle marbled is 4, PH24 is 5.95,intramuscular fat is 9.37%.A single muscle fiber cross-sectional area is 3498.333μ ㎡,fiber density is 256.25(root numbers/mm2).Compared with records,basic restore to its original level.Test two :This experiment studied seventy and 5-lineage shenxian breed hogs by using microsatellite DNA genetic markers.Polyacrylamide gel electrophoresis(PAGE)to test the genetic diversity by7 microsatellite loci(The United Nations food and agriculture organization(FAO) recommended twenty-three, choose seven).Using 7 microsatellite loci,measured many datas of each lineage,like Gene frequency,genotype frequency, heterozygosity, polymorphism information content and genetic distance, etc.The results of the experiment were shown as follows.shenxian pigs have a high reproductive performance,grow slowly, good meat quality and appearance also conforms to the variety characteristics.Breeds conservation of shenxian herds,53 alleles were detected at loci S0155,SW240,SW632,S0178,SW911,S0068 and SW857 with average 7.5714 alleles per locus.Especially SW240 at most,with 10.SW911 at least,with 5.According to the data analysis,we known that different lineages at the same loci have different allele frequency and allele number.so,for S0155,in 01 lineage with 6 allele,in 03 lineage with 5 allele,in 05 lineage with 5 allele,in 07 lineage with 4 allele,in 09 lineage with 2 allele,the numbers of alleles in each lineages were different from each other.This indicated that the genetic background of 5 lineages were different from each other.Besides,also analysed that the characteristics bands of shenxian pigs were found,can be made the herds genetic marker.The characteristics bands of line 07 were 94 bp band and line 03 were 116 bp band at locus SW240.At locus SW632,the characteristics bands of line 03 were 174 bp band.At locus S0178,the characteristics bands of line 07 were 138 bp band.At locus SW911,the characteristics bands of line 07 were 163 bp band.At locus S0068,the characteristics bands of line 05 were 238 bp band.At locus SW857,the characteristics bands of line 05 were 156 bp band.The range average locus heterozygosity of shenxian pigs 5 lineages was between 0.5873 and 0.7682.The locus average polymorphism information content was between 0.5309 and 0.7514. The result above showed that the genetic diversity of shenxian herds at these loci was high.According to the Nei’s standard genetic distance to understand the genetic relationship between families,genetic distance of 01 lineage,03 lineage and 05 lineage had a near relationship,commonly used in the production practice.
