Systematic and comprehensive research have been done on the regeneration of tomato and cucumber induced by several significant factors involve culture medium, hormone concentration and ratio of hormone concentration, which provides plenty of test reference and database for the further research on and bridge a gap between basic regenerative research and Genetic transformation. Lycopersicon esculentum Mill, Valley Floor Cucumber and Holland Cucumber Gulie168 were employed as the raw material in this study. The results are shown below: 1.Regeneration of tomato.Culture medium, hormone concentration and ratio of hormone concentration plays critical roles in modulating and regulating the callus induction, adventitious bud differentiation and root when the tomato was treating through cultured cotyledon & hypocotyl of Lycopersicon esculentum Mill into vitro, which have been Systematically researched, and Vc, an economic and environment-friendly reagent was found to have a significant and efficient function in inhibiting the browning phenomenon occurring in our study. The results showed that the optimum callus culture medium and hormone ratio was MS+(0.2-0.4)mg/L IAA+2.0mg/L 6-BA; The optimum medium to induct adventitious bud and hormone ratio was MS+0.1mg/L IAA+(1.0-3.0)mg/L 6-BA; The optimum medium and hormone ratio was 1/2MS+0.1mg/L NAA. Cotyledons was the best ex-plant because its better at this aspects:the speed & rate of adventitious bud occurrence and growth of the bud than hypocotyl, browning phenomenon could be efficiently controlled that added 10mg/L of Vc in callus induction medium. 2. Regeneration of CucumberBased on the experience and database summarized from the regeneration research of tomato. Valley Floor Cucumber and Holland Cucumber Gulie168, two universal cucumber varieties planted in china, were invested via a similar research method as tomato after optimizing the dose of disinfectants for sterilizing seeds. Mediated dose of AgNO3 was added and optimized to inhibit the vitrification of callus. The results of this research indicated that both of these two cucumbers can appear a great ability of callus differentiation. In contrast to tomato, Cotyledons is the best ex-plant, the hypocotyl acted a better callus differentiation ability than Cotyledons in these two cucumber, and the ratio of hormone concentration has a great potential to affect the callus’s morphological. In this work, we found out the best ratio of hormone concentration when Cotyledons were employed as the ex-plant is MS+0.5 mg/L 6-BA+0.1 mg/L IAA+1.0 mg/L AgNO3, while MS+2.0 mg/L 6-BA+0.1 mg/L IAA+1.0 mg/L AgNO3 were the best ratio of hormone concentration respectively for these two cucumbers when hypocotyl were employed as the explant. however, Adventitious buds did not show out in this condition, which need a further research.
