Chinese cabbage is a type of crop with seed vernalization, by feeling cold of germinated seeds to complete the vernalization. Premature bolting has serious influences on the products value of Chinese cabbage in spring cultivation, resulting in a great economic loss in agricultural production. FLC(Flowering Locus C) is one of the main genes influencing the vernalization requirement in the Chinese cabbage. In the present research, we used Chinese cabbage of monosomic addition lines introduced alien cabbage chromosome No.4 of different genotypes, translocation lines introduced cabbage gene Bo FLC3 and a ploidy series from one haploid as materials, expression of FLCs was performed by q RT-PCR, and the influence of FLCs on bloting was analysised. The main results were followed:1) The hybridizations between Chinese cabbage monosomic addition line AC4 and two Chinese cabbage inbred lines with early and late bolting were developed. The chromosome number of root tip of their offpsprings was identified, Chinese cabbage monosomic alien addition lines(2n=21) introduced by cabbage chromosome No.4 under different Chinese cabbage background were obtained. The result of bloting investigation for hybrid progenies from one cross combination indicated that the bolting of plants with chromosome 2n=21 was late than plants with 2n=20, and the bolting of progenies from diploid Chinese cabbage as male than that from diploid Chinese cabbage as female.2)The gene Br FLCs expression in haploid, diploid, aneuploid and tetraploid derived from one same genotype were analyzed by the real-time PCR. The results showed that the relative expression of each Br FLC gene was X>2X, 2.4X>4X during 0~7d vernalization treatments except for Br FLC5 with 2.4x<4x at 7d vernalization treatment. The relative expression of every Br FLC was the highest in 2X at 14 d vernalization treatment, and Br FLC2, Br FLC3 and Br FLC5 were X>4X. The relative expression of every Br FLC was X>2.4X, in which Br FLC2 and Br FLC3 was X>2X>4X at 21 d vernalization. The relative expression of Br FLC2, Br FLC3 and Br FLC5 were X>4X and 2X>2.4X at 28 vernalization treatment. The relative expression of Br FLC2 and Br FLC3 were X>4X, and Br FLC3 was 2.4X>or=4X at 5 different vernalization treatments.3)The relative expression of Br FLCs and Bo FLC3 were analysed by the real-time PCR in microspore culture plants from AC2. The results showed that the relative expression of FLCs gene was 23-4>6-1>23-3>6-3>4-1>23-7>6-2 at 0d vernalization treatment; The decreased degree of FLCs expression in microspore culture plants with Bo FLC3 were slower than that of plants without Bo FLC3 during 0~7d vernalization; During7~14d venalization, the decreased degree of FLCs expression in all of microspore culture plants were slower than that in 0~7d vernalization. The change endency in plants without Bo FLC3 was small rise first and then reduced, and in plants with Bo FLC3 it was small decreased or stabilized during 14~28d vernalization. The relative expression of Br FLC5 gene of microspore culture plants with Bo FLC3 was significantly lower than that of plants without Bo FLC3.
