Trollflower(Trollius chinensis) also known as Han Jinlian, which is Ranunculaceae Trollius annual or perennial herbaceous plants. At present, the research focused on trolliuschinensis Jinlianhua pharmacological component extraction, separation, identification and medicine preparation and quality control. As a better medicinal plants,Trollius has the stems and leaves of graceful shape, large showy flowers, which is also an excellent ornamental plants. Because of the high environmental requirements, it is rarely used as garden plants.The purpose of this paper is to compare the heat resistance of diploid plants and polyploid which was treated by colchicines,to provide the basis for solving the difficult problem that Trollius chinensis distributed in in high latitude areas were introduced to low altitude areas and most of them died in summer. Based on the earlier study, this paper made a further study to improve the method of Trollius polyploid survival rate and mutation rate, and determine the heat injury index, soluble matter content, membrane system damage, the protecting enzymes of antioxidant system activity, chlorophyll content, electrical impedance spectroscopy parameters to study the changes of physiological characteristics of the polyploid plants and diploid plant under heat stress. The results are as follows:1.The best disinfection solution of Trollius chinensis seeds : 75% alcohol disinfection for 5min and 0.1% Hg Cl2 disinfection for 10 min, and then continue to use 7.5% Na Cl O disinfection the seeds for 20 min. The ways of soaking seeds in 0.1% colchicine and 0.1% DMSO solution for 24 h and adding 0.1% colchicine solution into the culture medium can raise the mutation rate of Trollius chinensis plant, and The mutation rate was 19.67%, the variation rate of 17.33%.2.These phenomenons such as leaf spot, brown, wilting even death appeared in diploid plants were more than polyploidy plants,under 35 ℃ and 40 ℃. Under the temperature of 40 DEG C after 48 h treatment, the heat damage index of diploid plant was 80%, the heat injury index of polyploid was 66.67%,The polyploid plants have better heat resistance.3.The soluble matter content(the content of soluble protein and free proline content)of polyploid plants increased significantly compared with the diploid plants after high temperature stress. The damage degree of membrane system(MDA and the relative electric conductivity)of Polyploid plant was less than the diploid plants, the activities of antioxidant enzymes(SOD、POD)and the content of chlorophyll were more than the diploid plants. The electrical impedance parameter data like intracellular resistivity,extracellularandthe relaxation time of Polyploid plants was higher than that of diploid plants.4.Through the correlation analysis between the physiological indexes, the heat injury index, SOD activity, POD activity, intracellular resistance, relaxation time and distribution, which can be used as the main indicators to identify the thermotolerance of Trollius chinensis. The other physiological indicators can be used as the secondary indicators.5.The conclusion is that the heat resistance of the polyploid plants is better than the diploid plants by used of the method of fuzzy membership function to analyze the comprehensive evaluation of various physiological indexes. The semi lethal temperature of rollius diploid and polyploid plants were 35.34℃and 37.76 ℃ with EL method.The semi lethal temperature were 35.54 ℃and 38.79℃ with ri method.
