| Wanbei pig was a good local breed of lean meat pigs that had breeded by Anhui institution before 80’s in last century. The percent of raising the pigs once drove 68.98% in numbers of local breeds in Fuyang region. But now the pigs have been becoming extinct because of impolitic preserve for local breeds of pigs. As elevating of people’s living standard and increasing a demand of high-quailty meat of pigs in market, a economic value was renewed a realization for the pigs. However its hereditary basis has been changed enormously since it was breeding 20 years ago. So the germplasm resource of the pigs was studied in order to approach and uncover its characteristic, and furthermore to promote protecting and rationally exploiting the local breeds.The body weight and body size traits were investigated in 38 boars and 49 sows at 6 months old of Wanbei piglets, which could have instructional significance for conservation and utilization of local pigs. The selected piglets were weighted after 12-hour fasting at 0, 2,4,6 months old, and measured body length, body height, chest girth, chest depth and chest width at 3,4,5,6 months old, respectively. The results showed that average daily gain and absolute growth rate of all selected piglets were increased, and main body size traits were also gradually increased from birth to 6 months old. The weights of boars and sows at 6 months old were not significantly different (43.50±1.15 kg vs.41.05±1.26 kg) (P>0.05). The body length and chest girth of boars and sows during 4-6 months old were weak, body size traits were not significantly different between boars and sows (P>0.05).The peripheral blood cells were determined using automatic blood cell analyzer in 7 of healthy piglets selected at 3.5 months old and that of non-pregnancy sows at 4.5 years old of Wanbei pig, whose blood were taken respectively from their anterior vena cava and anticoagulated. The result showed that number of red blood cells (RBC) in peripheral blood and its relative other index in sows were more than in piglets, but no difference significantly between them (P>0.05) except mean corpuscular-hemoglobin concentration (MCC). MCC in sows was 317.25±1.55 g/L, that in piglets was 300.75±3.12 g/L, existed obviously difference (P<0.01). The peripheral blood leucocytes showed a descending tendency from adult to juvenile, and significantly difference between them in lymphoid leucocytes (P<0.05) and in white blood cell (WBC) (P<0.01). Compared with piglets, platelet number in sows decreased by 49.3%, was not significantly different (P>0.05).The peripheral blood cells were determined according to above method in 5 stock boars selected at 4 years old of Wanbei pig and Landrace respectively. The result showed that an ascension were observed in RBC, hemoglobin concentration (HC), volume of packed red cells (VPRC), blood dick, thrombocytocrit and etc. of Wanbei pigs selected for the experiment, compared with Landrace, but it was not significantly difference (P>0.05). The number of WBC and its leukocyte differential count was not significantly difference between two breeds (P>0.05).The traits of piglet born were investigated in 35 pregnancy sows (9 primiparas and 26 multiparas) of Wanbei pigs and 38 sows (14 primiparas and 24 multiparas) of Landrace selected for the experiment. The result showed that the litter size were averagely 9.92±1.15 and 10.83±0.56 respectively in primiparas and multiparas of Wanbei pigs, and decreased by 18.04% and 8.40% compared with that of Landrace (P>0.05). The litter piglets alive born were droped by 13.49% in primiparas of the former breeding, lower than in the latter (P>0.05). But in the multiparas the number increased by 5.12%, not observed obviously difference between the two breeds (P>0.05).Genetic distance were analyzed between individuals among Wanbei pigs by random amplified polymorphic DNA (RAPD) and using L (35) orthogonal design. The result showed that the optimal RAPD reaction system was 3.00mmol/L Mg2+,1.50U Taq DNA Polymerase,0.40mmol/L dNTPs,0.50μmol/L Specific primers, lmg/L DNA template.64 cDNA visualized in the agarose gel electrophoresis were amplificated by the RAPD reaction system from 20 pigs, and those size of molecular weight located 0.20 kb-2.50 kb. The number of polymorphic DNA was 31, its percentage 48.44%. The max in similar rate*" of cDAN was 0.970±0.005, and that minimum was 0.870±0.018. The rate of heterozygosis was 0.113 averagely.The conclusion showed that average daily gain and absolute growth rate of piglets were gradually increased in Wanbei pigs from birth to 6 months old, and absolute growth rate of boars was higher than that of sows. However, the body weight and main body size traits were not significantly different between the boars and the sows. The functions in immunization and transportation of O2 and CO2, were perhaps stronger in the adult sows than in the piglets. The fluctuation in number of blood plaques within a physiological extent was intensified more in the piglets than in the adult. No difference existed both in litter size of sows and in number of WBC and of blood plate and related indexes of boars between Wanbei pigs and Landrace. The fluctuation in number of blood plaques within a physiological extent was obviously more in Wanbei pigs than in Landrace selected. The genetic state of Wanbei pig herds which had been collected again are disequilibrium and no clear relationship between the individuals. |
