BNS was bred by Professor Ru in Henan Institute of Science and Technology. Northwest A&F University fond commen wheat line 9833 is a restoring line for BNS. It has broad application foreground. We construct the F2 population of BNS and 9833 to construct the molecular genetic linkage map, the genotype identification was taken by SSR molecular markers, based on the method of PCR, the molecular genetic linkage map was constructed with software Ici Mapping3.2, and the QTL analyses were performed with software Ici Mapping3.2 too. The results are as follows:1 The survey of seed setting rate and genetic analysis of parents, F1 and F2 population showed that he fertility restoration of 9833 is controlled by 2 major genes, conforming to the genetic model of 2MG-AD-AD.2 Taking the F2 population as mapping population, the genetic linkage map was constructed by SSR molecular markers. Composing 17 linkage groups, including 84 couples of SSR primers. The total genetic length is 2444.13 cM, the average interval distance is 29.09 cM. The number of SSR primers on chromosome Aã€Bã€D are 31ã€29ã€24, severally covering 711.76cMã€798.83cMã€933.54 cM.3 QTL analysis of restoring gene in 9833, detected 4 QTLs(LOD>3), naming QRF-1A-1, QRF-1D-1, QRF-1D-2 and QRF-5A-1, QRF-1D-2 and QRF-5A-1 are the major QTLs, QRF-1D-2 located on chromosome 1D, from Xwmc216 to Xgdm111 and QRF-5A-1 located on chromosome 5A, from Xgpw7007 to Xbarc186, explaining 22.93% and 19.56% percentage of the phenotypic variation. The other 2 QTLs located on chromosome 1A and 1D. The 4 QTLs are different from the QTLs that had been reported. |
