Sarafloxacin hydrochloride belong to quinolones third-generation fluoroquinolones, a class of dedicated animal veterinary products. The Domestic drugs in animal clinical applications increasingly widespread, with the use of the drug appears the drug resistance and animal tissue residue etal. In this paper, 5% sarafloxacin hydrochloric solution of veterinary medicine production enterprises in test samples of research for the study, detection the drug stability under accelerated conditions, the minimum inhibitory concentration and residues studies of remains in the edible tissue of chickens, obtain the following results. This paper will test samples of 5% sarafloxacin hydrochloric solution accelerated into the chamber by a commercial package, at a temperature of 40 ± 2 ℃, relative humidity 70% ±2% under the conditions of five months, sampling 1 month times. Analyte the concentration of the drug measured by HPLC. Mobile phase: 0.05 mol / L citrate-0.05 mol / L ammonium acetate- acetonitrile(80: 10:18), with perchloric acid to adjust the p H to 2.4; detection wavelength was 274 nm, using the Agilent 5TC-C18(2) 4.6 × 250 mm, 5 μm column. The test samples are compared with the results of the acceleration detected by the front, little change in the first three months of drug content, assay results are 99.8%, 99.2% and 100.2%, after two months, a slight degradation of the drug content but the change in scope, the found of content to be 97.9% and 94.5%. Two months after the drug content is slightly reduced, but not been found the chromatogram(descending) hydrolyzate. The study sample of 5% sarafloxacin hydrochloric solution vitro antibacterial effect, compare the minimum inhibitory concentration of 6 quinolones of quality control strains of E. coli, E. coli clinical isolates, Salmonella clinical isolates.Drugs were diluted to 1280 μg / m L, add broth using 96 serial dilutions, dilution to turn the first nine holes, the first 10 holes plus 100 u L liquid medication as a drug control, the first 11 holes plus 100 u L MH broth as a negative control. The results 5% sarafloxacin hydrochloric solution of the three strains have strong antibacterial activity, quality control in E. coli strains and clinical isolates of E. coli in a strong antibacterial activity, clinical isolates of Salmonella compared to other similar strains of bacteria sensitive to drugs is relatively weak. The study of Sarafloxacin residue fed to broiler recommended dose of 10 mg / kg, was detected in liver, kidney, muscle tissue residue concentrations of the drug. Mobile phase: V(0.02 M p H3.5 sodium dihydrogen phosphate): V(acetonitrile) = 82: 25, flow rate 1 m L / min, detection wavelength: 274 nm, the column was Agilent 5TC-C18(2) 4.6 × 250 mm, 5 μm. After 24 h discontinuation the drug residues in chicken liver and kidney tissue sarafloxacin content lower than the national standard MRLs, residues in muscle tissue in 48 h higher than the national standard MRLs.In conclusion, 5% sarafloxacin hydrochloric solution have a better stability, low minimum inhibitory concentration, eliminate rapidly in chicken liver and kidney tissue, muscle tissue slowly.
