Research On The Minimum Inhibitory Concentration In Vitro Of Sticktight On Esbls E.Coli

MIC of common antibiotics against16clinical Escherichia coli from Colibacillosis in pig were determined by the ameliorative two fold dilution method,the ESBLs produced by Escherichia coli were detected by double slips synergy,and using PCR method to make sure its genotype. It showed that the bacteriostatic effects of traditional Chinese medicine against the drug-resistant strains.The results showed that in all drug-resistan stiains,The rate of ceftriaxone and amoxicillin resistance was100%;and observed that resistant to ciprofloxacin, norfloxacin, enrofloxacin(87.5%), the rate of Levofloxacin and gatifloxacin resistance was25%,and50%drug-resistant strain were sensitived;the ESBLs-producing strains have high resistance rate to mequindox,amikacin,lincomycin,colistin,doxycycline,ceftazidime, while the resistance rate to these agents was62.5%,62.5%,43.1%,37.5%,75%,75%, and56.3%separately. ESBLs producing results show that all drug-resistant strains contain resistance genes,TEM genotype detection rate of100%,SHV gene detection rate of12.5%,CTX-M genes detection rate of43.8%,the detection rate of OXA genes31.3%.Meanwhile there were13strains containing two genes accounting for80%drug-resistant strains,moreover there were2strains containing three genes.Test results prove that Bidens Bipinnata was highly effective against ESBLs produced of Escherichia coli.Its MIC was0.5g.ml-1. Antibacterial experiments in chessboard method and passaging in Chinese and Western medicine showed that amoxicillin,ceftazidime, ceftriaxone, cefodizime, mequindox, amikacin, doxycycline and Bidens Bipinnata liquid boiled have combined antibacterial effect.In different solvents extract combined antibacterial effect, ethyl acetate extracts> boiled liquid> acetone extract> chloroform extract.The plasma concentration-time data derived from experiments were analyzed by non-compartmentalmethod based on statistical moment theory. The main pharmacokinetic parameters after intravenous administration were as follows: AUC(108.2) mg·h·L·1, ClB(1.480) mL·h-1·kg-1, Vz/F (18.37) ml.kg-1, t1/2λ (11.29±1.40) h, Cmax (3.32±0.33)ug.ml-1,Tmax (1.13±0.44)h.
