Ralstonia solanacearum, one of the most widely world distributed and economically important plant pathogen, has been studied by the ion exchange high performance liquid chromatography (IE-HPLC) to setup identity for recognition of R. solanacearum stains. By chromatographic analysis of strains from different sources, the study on pathogenicity of R. solanacearum strains in different parts of plants in different host showed the different chromatographic behavior, based on which some of the strains such as Tn-5 inserted R. solanacearum strains’explored the colonization ability and biological control effect in tomato bacterial wilt disease. The relationship between biological efficacy and chromatographic behavior was to be studied to screen the titer index for plant vaccine. The results showed as follows:1 Virulent strains of R. solanacearum were recognized with attenuation index less than 0.65, while the avirulent strains with attenuation index higher than 0.75. The study determined attenuation index of 71 strains of Ralstonia solanacearum strains which from different sources. Those R. solanacearum had the index between 0.65-0.75 were considered as interim strains. Based on this research, there are 71 Test strains, which 29 strains of pathogenic strains of strong,8 strains of transition and 34 strains of avirulent strains.2 To optimize Separation and characterization conditions of R. solanacearum by using chromatographic. (1) To set different PH (6,7,8,9), the impacts of balance and elution buffer was analyzed under different PH conditions. (2) To set different sample size (10 μl,20 μl,30μl,40 μl,50 μl), and impacts of different sample size were analyzed. The best condition of chromatographic of R.solanacearum was pH7.0 and sample size 20uL.3 Two different pathogenicity strains FJAT-91 and FJAT-1458 are characterization by chromatographic as models to distinguish 71 straina of different virulence of R. solanacearum strains by the characteristics. According to the characteristics of their peak numbers, these 35 strains of bacteria were divided into 3 categories. The first group, appeared a single characteristic peaks in lmin of retention time, the second goup, two peaks in lmin and 8min of retention time, the third class, three peaks in lmin,8min and 17min of retention time. The analysis of characteristic spectra indicated that the different pathogenicity strains had different peaks and retention time in the chromatography.4 To compared colonization characteristics and prevention effects of twenty-nine insert-attenuated EZ-Tn5 TM strains, constructed chromatography titer index of mutant strains, and analyzed the correlation between chromatography titer index and biocontrol efficacy. The results showed that all of the twenty-nine strains could be colonizated in potted tomato seedlings; the longest colonization time was 20 d. Strain T871 is the best colonization strain, after 20 d, the colonization biomass reached 2.8 ×104 cfu/(g FW); while Strain T713 and T831 have the best control for bacterial wilt of tomato, the proportion of prevention reached 91.7%. Through correlation analysis, the relationship between chromatography potency (X1) and biological control (X2) was positively related, the correlation coefficient is 0.60.
