Since the early coastal shelterbelt construction in the content and structure of a single,resulting protective effect gradually weakened,the southeast coastal areas of Fujian currently has clumping bamboo and other large coastal shelterbelt species introduction as greatly improved the ecological structure of coastal protection forest,where bamboo is One of bamboo species better adaptability to study the mechanism of salt tolerance is particularly important breeding strains Salt current research is conducted from bamboo salt stress on physiological and biochemical level,but not related to bamboo Salt-related class proteomic level,has not been found on the bamboo salt stress differential proteome research reports.In this study,Fujian major science and technology(2013NZ0001-l)coastal sandy project,supported by the introduction of bamboo,combined with the results of previous studies,Zhangzhou City,Fujian Coastal Sandy Land Farm Chishan introduction bamboo as research material,through asexual Department of bamboo material acquisition,construction of leaf protein electrophoresis system,the salt stress response protein isolated and identified by mass spectrometry to study the type and function of proteins in response to explore differences bamboo salt stress protein response mechanism,and then from the molecular level changes when bamboo salt stress protein expression from the gene level to provide a theoretical basis for the bamboo salt tolerance and salt tolerance breeding of new varieties.In the study of clonal bamboo material obtained through eight orthogonal group bamboo cuttings processing,statistical rooting rate,analysis results applied to bamboo cutting seedling cuttings program :complete bamboo cuttings collateral material selection with pocket portion,stay 3-4,to leaves,soaked overnight after water solution with 500ppmABT ABT hormone treatment,after treatment to sandy peat cuttings branches mixing matrix.Two-dimensional electrophoresis system build bamboo leaf total protein research,access to green bamboo leaf applied to two-dimensional electrophoresis system:protein samples using TCA/acetone+SDS phenol extraction emulated get,like the hydration of the 17cm pH3-10 IPG strips passive hydration 24 hours,when the sample strip has been adopted on the IEF gel aggregates oriented mode,the focus program is set to 80000Vh,obtain reproducible,high resolution gel images.After clonal bamboo seedlings were 0.3% and 0.5% NaCl treatment for two weeks on leaves of two-dimensional electrophoresis protein,0.3%concentration in the control group and the experimental group were measured 895,884,841,754,786,738 months proteins;0.5% concentration in the control group and the experimental group were measured 980,953,942,762,715,730 protein spots.0.3%mild stress induced in response to salt stress found 394 protein spots:36 new protein spots,seven abatement spots,213 regulated protein spots,138 down-regulated proteins.Significant differences in the selection of six points in MALDI-TOF-MS detection,discovery and rice functional proteins homologous protein TRIUR308890 Os02g0598300 and assumptions for the new protein,a class millet DNA primase large subunit,triosephosphate isomerase in salt stress induced upregulation of class millet DNA mismatch repair protein MSH7,tRNA dimethylallyl transferase downregulated in salt stress induction.0.5%severe stress induced in response to salt stress found 338 protein spots:115 new protein spots,73 protein spots were cut,93 protein spots were up-regulated and 57 down-regulated proteins.Cut nine more significant differentially expressed protein spots were MALDI-TOF-MS detection,discover unknown loc100276336,70 kDa heat shock protein-related protein,argonaute 1B protein,glyceraldehyde-3-phosphate dehydrogenase 2,Rubisco large subunit a subunit protein upregulated in response to salt stress,hypothetical protein osi20531,ATP synthase,TIR-NBS-LRR class resistance protein down-regulated in response to salt stress.The study found that 70 kDa heat shock-related protein and Rubisco large subunit binding proteinasubunits in the bamboo salt are highly expressed in response to stress,they assume important functions in bamboo salt resistance mechanisms.And find TIR-NBS-LRR class resistance protein is the first time in bamboo leaf protein.
