The popularization and application of a large area of hybrid rice has made a great contribution for food security in our country. To ensure the purity of hybrid rice seeds more than 96% is a key indicator of hybrid rice seed quality. Traditional hybrid rice seed purity testing methods is to use the field planting in natural greenhouse conditions of the winter in Hainan, observed differences in rice plant traits to determining seed purity, the identification method takes about three months time, unfavorable for seed sales, and also affected the rapid adoption of new super hybrid rice combinations. Therefore,establish a rapid and accurate identification method of hybrid rice seed purity is an urgent demand for hybrid seed industry. In this study, using zhu1 S,Y58S such as 36 dual-use sterile lines rice as materials to establish SSR markers library, on this basis, screening male and sterile lines characteristic molecular markers to construct rapid methods for hybrid seed purity identification, aimed at promoting the development of hybrid rice seed industry.The main conclusions are as follows:1. For defects that national agricultural industry standards recommended SSR markers RM274 located on the 5th chromosome polymorphism is poor, we developed polymorphic RM163. And wealso recommend using the RM163 replace of RM274 during DNA fingerprinting analysis of new varieties.2. We using the result of 24 pair primers in national standard and newly developed primers RM163 build 36 rice varieties DNA fingerprinting which been processed into a form of two-dimensional code that computer can recognize, analyze and calculate.3. In this study, we build Dual-use sterile line two-dimensional fingerprint and SSR markers library which can be successfully used to identify hybrid seed purity, using RM71 can identified the hybrid seed purity of ZY series, the application of newly development primers RM163 can identified the seed purity of GZ63 hybrid combination series,which can effectively distinguish the doped seed from true seed.The seed purity of three ZY series varieties all less than 96% are unqualified varieties. In GZ63 hybrid combination series : The seed purity of GZ63/R96 was 96.82% which is qualified varieties, and GZ63/R25, GZ63/R63, GZ63/R42 less than 96% are unqualified varieties.
