Traditional Chinese herbal medicine isindispensable part in Chinese medicine.A bulk of studies have confirmed that many herbal medicines and their components have the function of enhancing immunity, anti-oxidative, anti-tumor, and many other effects. In this study, the immune and anti-oxidative function of Chinese herbal formula WQB(Wei Qi Booster) on aged mice was analysed, and the acute and long-term toxicity of WQB on young mice is studied to provide the theoretical basis in clinic.This study was carried out to investigate the effects of WQB on immune parameters and anti-oxidative function in aged mice. Fifty mice were randomly divided into five groups. Group A was kept as control group. Mice in Group B were receiving10mg/kg body weight(BW)Levamisole. Each mouse in groups C, D and E was received 0.1%, 1%, 2%WQB respectively. Another 10 young mice were fed regularly as group F. The mice were fed for 30 days according to the above methods. Results showed that compared with the control group, the body weight and immune organs indexes were significantly enhanced in the group with 1% WQB(P<0.01). In addition, 1%WQB could improve the activity of SOD and IL-2, while the content of MDAdecreased(P<0.05, P<0.01). The positive cells of CD4 increased while the levels of CD8 decreased in jejunum, and the expression of s Ig A was increased.IFN-γ contents in the 1%, 2% WQB group increased, and there is no significant difference compared with the young mice group. Results showed that WQB can increase the immune level and enhance the anti-oxidative function in aged mice.The acute toxicity test was conducted to obtain the safety data of WQB. Eighty Kunming(KM) mice were randomly divided into 7 WQB groups which drench 2ml/kg BW,2.80ml/kg BW,3.92ml/kg BW,5.49ml/kg BW,7.68ml/kg BW,10.76ml/kg BW,15.06ml/kg BW respectively and a control group which were feed normal saline. No death were observed in 7 WQB groups after 7 days treatment. The maximum tolerated dose(MTD) of WQB was 30.12ml/kg BW, which equal to 0.9g per mouse. After treatment, no death were observed. The weight of liver and kidney were calculated and the liver, spleen, lung, kidney and duodenum were fixed for morphology observation through hematoxylin-eosin(H.E). Besides, the haematological traits eg. Intermediate cell(MID), Lymphocyte(LYM), Red blood cell(RBC), Hemoglobin(HGB)were measured. Results showed that there were no significant differences of the weights in liver and kidney at 0.05 and 0.01 level and no adverse effects on organs morphology among the groups of 15.06 ml/kg BW WQB, the MTD and the control of acute toxicity test. After herbal administration, there were no correlation in haematological traits between the WQB groups, MTD group and control group(P>0.05).The chronic toxicity test was carried out to get the safety data of WQB.In the present test, fifteen mice were divided into three groups as control group, 10mg/kg BWLevamisole group and WQB group. The mice in the control group were fed as normal, mice in the positive group were fed10mg/kg BW Levamisole,mice in WQB group were fed with 1% WQB.After 90 days treatment. The weight of liver and kidney were calculated and the liver, spleen, lung, kidney and duodenum in control group and 1% WQB group were fixed for morphology observation through H.E. Besides, the biological indexeseg. Triglyceride(TG), Cholestrol(CHOL), Glucose(GLU), Uric Acid(UA), Creatinine(Crea), Glutamic-pyruvic Transaminase(ALT), Blood Urea Nitrogen(BUN), Total Protein(TP), Albumin(ALB) and Aspartate Transaminase(AST)were measured. Results showed that there were no significant differences of the weights in liver and kidney and no adverse effects on organs morphology between the 1% WQB group and the control group of chronic toxicity. Moreover, after herbal administration, there were no statistically significant difference of biological indexes in the two groups(P>0.05).It is conclude that 1% WQB can be used as an immune enhancing agent and anti-oxidant with reliable safety.
