Pseudomonas aeruginosa is a common zoonotic opportunistic pathogens, brings a great threat to human and animal health. 51 bacterial strains were isolated from clinical in this study, and their resistant phenotype, resistant genotype, exotoxin and pathogenicity of animals were detected and analyzed, providing basic data for clinical Pseudomonas aeruginosa disease research.Since2011-2014 in He Bei and its around areas, a total of 51 wild virus isolated and purified from clinical onset chickens and swines, which identified by morphology and PCR technology. Using Kirby-Bauer disc diffusion method(K-B method), antimicrobial susceptibility tests to 3 categories of 17 kinds on antibacterial agents, performed multiple drug resistance. The results show that the rate of resistance against aminoglycoside were as follows: streptomycin: 84.3%, amikacin: 84.3%, kanamycin: 78.4%, neomycin: 78.4%, gentamicin: 54.9%, spectinomycin: 25.2%. The rate of resistance against quinolone were as follows: ofloxacin: 94.1%, enrofloxacin: 82.4%, levofloxacin: 80.4%, gatifloxacin: 78.4%, norfloxacin: 62.7%, ciprofloxacin: 25.2%. The rate of resistance against β-lactam were as follows: cefradine: 98.0%, penicillin: 98.0%, ceftriaxone: 70.6%, cefotaxime: 2.0%, cefoperazone sodium / sulbactam sodium: 0.0%.In this paper, 51 P. aeruginosa were tested for the corresponding resistance genes using the polymerase chain reaction(PCR). The results of 6 kinds of aminoglycoside resistance gene(AMEs) were as follows: the positive rates of ant(3′′)-Ia, aac(6′)-Ib, aac(3)-IIa, aph(3′′)-Ib(str A), aph(6′)-Id(str B), aph(3′)-IIa from high to low were 98.0%, 51.0%, 43.1%, 41.2%, 31.4%, 17.6%; one of AMEs was negative, three of AMEs were whole carriers. The test results of 16 S r RNA methylases(arm A, rmt A, rmt B)were negative. The positive rates of quinolone resistance plasmid from high to low were:aac(6′)-Ib-cr(13.7%), qnr S(9.8%), qnr B(5.9%). Quinolone-resistant determining region(QRDR) of gyr A gene were detected by PCR, sequenced and the sequence analysis. These results indicated that the QRDR gene mutated on 83 amino acids only occur in three bacterial strains: 83Thrâ†'Ile. The positive rates of β-lactam resistance gene from high to low were OXA-3(7.8%), OXA-1(2.0%), OXA-5(0.0%).The results of PCR test show: the positive rates of 51 P aeruginosa isolates cytotoxic exotoxin A(ETA) were 72.5%. Animal pathogenic regression experiment proves that 51 P. aeruginosa isolates have strong pathogenicity. Key words: Pseudomonas aeruginosa; Drug resistance; Resistance gene; Separation and...
