The development of proteomics technology provides a new perspective and method for functional genomics research. Researching goat skin hair follicle development from the perspective of proteomics and finding target protein by comparing differences protein is a new way to study the goat breeding. As Keratin family widely expressed in the hair follicles, while they are also the main components of the cashmere, it may be the important regulatory factors of the hair follicle Cycle. In this study,we analysis skins from the cashmere growing in anagen and telogen by using iTRAQ and LC-MS/MS. Proteins were quantitated and identificated. Differences proteins are classed by GO Term and KEGG PATHWAY analysis. The proteomic profiling of goat skin and different expresstion proteomic profiling were constructed for the first time, which lay a foundation for functional genomics research. The main results wereas follows:1.724 protein from goat skin was identificated,721 identificated protein are high reliable proteins wich can be mapping at less two peptides.44 Keratin was identificated,wich account 6% of total identificated protein.2. Identificated protein was analysised by classied in Cellular Component, Molecular Function, Bigological Process three aspects:there are nine kinds proteins in Cellular Component, ten kinds proteins in Bigological Process, eighteen kinds proteins in Molecular Function.3.211 KEGG pathway was mapped.by total protein in this KEGG enrichment analysis.4. iTRAQ technology are used to get the 158 differentially expressed proteins.72 proteins are up-regulated expression and 86 proteins are down-regulated expression.9 Keratin proteins are up-regulated expression,2 Keratin proteins are down-regulated.5. Differences proteins were analysed from Protein-Protein Interactions net work by string 10. A Protein-Protein Iteractions net composed of 40 protein was got.6. The result of quantitated by iTRAQ was identificated by Western Blot.
