In this study, the full sequence of Wuzhumuqin sheep MSTN gene was cloned and sequenced. The sequence structure, composition of nucleotide, and encoded amino acid were analyzed by bioinformatics, and the molecular evolutionary tree was established. Furthermore, the polymorphism of the partial nucleotide sequence of Wuzhumuqin sheep MSTN gene was analyzed by PCR-SSCP.The results shows that Wuzhumuqin sheep MSTN gene had 3 exons and 2 introns. The size of exon â… , exon â…¡ and exonâ…¢ was 373bp,374bp,381bp. While the size of intron I and intron â…¡was 1833bp,2030bp. The size of CDS sequence of Wuzhumuqin sheep MSTN gene was 1128bp, encoding 375 amino acid. Biological software analysis showed that the theoretical molecular weight of Wuzhumuqin sheep MSTN gene was 42.8 kDa and PI was 7.01. Leu was the highest (9.9%), followed by Lys (8.3%). The secondary structure of Wuzhumuqin sheep MSTN protein was mainly Random coil and it had characteristics of transmembrane protein structure and the structure located between AA6-AA23. It also had a secretion signal peptide structure. The anino acid sequences MQKLQICVYIYLFMLIVA, which related well with TGF-β family. One mutation site was detected in the P6 primer amplified 366bp sequence of MSTN gene in Wuzhumuqin sheep, Only one Câ†'T transition was found at this region, it is a synonymous mutation site, and formed 2 genotypes, named EE and EF. There was no polymorphic site detected in the P7 primer amplified 353bp sequence of MSTN gene in Wuzhumuqin sheep. One mutation site was detected in the P8 primer amplified 349bp sequence of MSTN gene in Wuzhumuqin sheep, Only one Aâ†'G transition was found at this region, it is a synonymous mutation site, and formed 2 genotypes, named AA and AB. The study will provide reliable information on the establishment of wuzhumuqin sheep gene library, and also provide the theoretical basis for the improvement of local sheep breed and the study on the usage of MSTN gene as a molecular breeding technology of wuzhumuqin sheep.
