Myostatin(or GDF-8), a member of TGF-β superfamily, is negative regulator of skeletal muscle mass.SNPs (single nucleotide polymorphisms) is the most important research method in Molecular Genetics. Firstly ,the PCR-SSCP with about 300bp fragment have been established in the experiment.And then the present study selected MSTN as a candidate gene to identify any difference in 2512 bp fragment included one intron and three extrons of MSTN among Wushen, Wuzhumuqin, Xinihe, Sanhe, Thorougnbred and Short Horse by PCR-SSCP, PCR-RFLP. As a result of PCR-SSCP,the transversion of G→A in 95th and A→G in 337th of first intron cDNA lead to Amino acids mutation of AGT→AAT and ACG→GCG. The transversion of T→C in 809th of third intron cDNA result in Amino acids mutation of CTT→CCT. As a result of PCR-RFLP, there is not the restriction site of SrfI, and polymorphisms after cutting with ScaI,MspI and HhaI. However both restriction site and polymorphisms exist after cutting DraⅠ.
