In this paper, we used Inner Mongolia cashmere goats as experimental model and performed RNA-seq to three tissues (hypothalamus, pituitary, muscle) transcriptome libraries of two groups cashmere goats (group A:BMI≥90, group B:BMI≤80). Then, we explored the different expressed genes (DEGs) between the two groups and performed GO functional annotation and KEGG pathway analysis for the DEGs to certain which most important biological metabolism pathway and signaling pathway they may involved. These results provided theoretical basis for the regulation mechanism of obesity in cashmere goat. The main results are as follows:1. We successfully constructed 18 transcriptome samples libraries of hypothalamus, pituitary, muscle in 6 Inner Mongolia cashmere goats. Based on RNA-seq high-throughput sequencing technology, We totally obtained 68GB expression profile data.2. Comparison of different expression genes in group A and group B, the results are as follows:In the hypothalamus, a total of 4900 differentially expressed genes were obtained including 4398 up-regulated genes and 502 down-regulated genes; In the pituitary, a total of 721 differentially expressed genes were obtained including 298 up-regulated genes and 423 down-regulated genes; In the muscle, a total of 686 differentially expressed genes were obtained including 276 up-regulated genes and 410 down-regulated genes.3. The analysis of Venn diagram of differentially co-expressed genes, the results are as follows:196 differentially co-expressed genes were obtained in hypothalamus vs. pituitary; 170 differentially co-expressed genes were obtained in hypothalamus vs. muscle; 48 differentially co-expressed genes were obtained in pituitary vs. muscle; 33 differentially co-expressed genes were obtained in hypothalamus vs. pituitary vs. muscle.4. KEGG pathway analysis of differentially expressed genes, the results are as follows:①The differentially expressed genes in hypothalamus were mainly involved in focal adhesion, axon guidance, regulation of actin cytoskeleton, ECM-receptor interaction, oxidative phosphorylation, TGF-β signaling pathway, phosphatidylinositol signaling system, ErbB signaling pathway, MAPK signaling pathway, calcium signaling pathway, insulin signaling pathway, Notch signaling pathway and Fc gamma R-mediated phagocytosis. The differentially exp ressed genes in pituitary were mainly involved in neuroactive ligand-receptor interaction and tight-junction. The differentially expressed genes in muscle were mainly involved in focal adhesion, ECM-receptor interaction, insulin signaling pathway, alanine,aspartate and glutmate matabolic, Phenylalanine, tyrosine and tryptophan biosynthesis, adipocytokine signaling pathway, cell adhesion molecules (CAMs) and Glycolysis/Gluconeogenesis.② The differentially expressed genes in hypothalamus vs. pituitary were mainly involved in Cytokine-cytokine receptor interaction; The differentially expressed genes in hypothalamus vs. pituitary were mainly involved in ECM receptor interaction and fructose and mannose metabolism; The differentially expressed genes in piyuitary vs. muscle were mainly involved in chemokine signaling pathway.5. AQP1, ADAMTS2, UBE2L6, ZBTB16, ECM receptor interaction and chemokine signaling pathway might be play an important role in the obesity regulated by hypothalamus-pituitary-muscle axis.
