| Inner Mongolia Cashmere goats (IMCGs) are valuable germplasm genetic resources because of having many excellent cashmere production traits. So it is particularly important to study IMCGs’hair growth mechanism and explore an important candidate gene that associated with the hair growth. In this test, we selected three different periods (Anagen in Novemberã€Catagen in Januaryã€Telogen in March) of Inner Mongolia Cashmere’s primary follicles and secondary follicles from side for three RNA samples and mixed them together. Then used High-throughput Sequencing Technology to sequence transcriptome and analyzed the results by bioinformatics and q RT-PCR verification. Test results showed:1. Filtering primary follicle and secondary follicle between groups and within groups in three different growth periods, we found nine genes were differently expressed. Analyzed these nine genes by GO and Pathway analysis. GO analysis showed:Most significantly enriched entries are related with extracellular matrix organization, structural constituent of ribosome, translational elongation, cytosolic small ribosomal subunit, actin binding and vitamin K metabolic process. Pathway analysis showed that these different genes are significantly worked on ribosome, amino acid metabolism, vitamin metabolism, sulfur metabolism, neomycin biosynthesis, etc.2. The test had the genetic screening within groups in different periods of primary follicle and secondary follicle, we found that these genes in three periods (anagen, catagen, telogen) has eight kinds of continuous change in trend:down down down, up up down, down up down, down up up, up down down, up down up, down down up, up up up, and the first three trends are more significant. Analyze the different genes in these trends will provide a strong basis in digging follicle (especially secondary follicle) cyclical growth control mechanisms.4. Real-time quantitative PCR was employed to quantify the LGR5 and SOSTDC1 expression in Cashmere goat hair follicles. The expression of LGR5 and SOSTDC1 in November was significantly higher than that in March. This is consistent with the results of the sequencing. The result of sequencing is real and effective. |
