Inner Mongolia cashmere goat is one of the rare species of China, its cashmere fiber has high economic value which is known as the "diamond fiber". Previous studies found that melatonin can promote the growth of hair follicle, but the signaling pathway and regulation mechanism are not clear yet. Therefore, we took Inner Mongolia cashmere goats as research object, implanted melatonin in vitro, used Western blot and immunofluorescence technology, to research the protein expression of RORa in Inner Mongolia cashmere goats in February, September, and December.The results showed that:(1) The growth characteristics of cashmere and the test wool quality results showed that the difference of fineness of cashmere of September and December between the experimental group and the control group is significant (P<0.05), other months were not significantly different (P>0.05); The length of cashmere of experimental group and the control group in September, October, November, December, January, February, and March was significantly different (P<0.05) from each other, and it was not significant (P>0.05) in August, December and April.The results showed that melatonin has a trendency to promote hair to become longer; The difference between the experimental group and the control group was not significant, but there was a trend of increasing the yield. The results showed that MT had a great influence on the length of the villi, and had little effect on the fineness of the wool and the yield, but there was a trend of increasing the yield.(2) Western blot results showed:the expression of RORa proteins of the experimental group and control group was found in hair papilla. The expression of RORa proteins in February was highest, decreased in September, and lowest in December. The expression of RORa in the experimental group was higher than control group (p<0.05),while in September and December there’s no significant difference (p>0.05).The results showed that melatonin improved the expression of RORa in February.(3) Immunofluorescence staining results showed that in the control group,RORa only expressed in the outer root sheath of the hair follicle in February, September, and December. The expression of RORa in the experimental group protein was fond in the root sheath of the hair follicle and hair papilla in the three months.This experiment works on the influence of implanting melatonin on the growth characteristics and the quality of cashmere. We used Western blot and immunofluorescence to study the expression of RORa in the Skin hair follicles of cashmere goats after implanting melatonin. The results assumed that melatonin combined with RORa which express on the root sheath of hair and hair papilla to influence the growth of hair, and involved in adjusting the growth circle of hair follicle.
