The masson pine(Pinus massoniana Lamb.) is one of the main afforestation species which is widely distributed in China. In recent years, the masson pine has been severely damaged by pine moth(Dendrolimus punctatus) which has caused huge amount of economic loss. Therefore protecting Pinus massoniana forest is becoming increasingly critical.At present,there are many reports about comparison between damaged and undamaged pine needle.Few involves comparison of insect-resistant and non-resistant pine needle.In order to investigate the difference between undamaged insect-resistant and non-resistant pine needles and the seasonal variation of the insect resistant clones,we studied the chemical and physiological differences among insect-resistant masson pine clones GK001, GK002 and control clones.Each clone needle was sampled repeatedly on late March,late May,late August and late October.Content of soluble sugar,content of soluble protein,SOD activity,POD activity were determined.With method of variance analysis, monthly differences of physiological and biochemical parameters of pine needle were determined. Volatile metabolite content of pine needle has been quantified by using gas phase chromatography technology. Gas chromatography and mass spectrometry GC-MS was used to qualitify volatile secondary metabolites in needles of Pinus massoniana. Variation of volatile secondary metabolites content among month and variation between insect resistant clone and controls were tested by variance analysis.The main conclusions are as follows:1.Content of soluble sugar of needle from insect resistant and control masson pine clones changes in a similar trendency and it was detected differences among months; Content of soluble protein of needle from insect resistant and control masson pine clones change in a similar trendency and it was detected differences among month. Insect resistant and control masson pine clones have similar changing tendency of SOD, POD activity of needle and there exist differences in SOD activity in March and May between insect resistant and control masson pine clones; there exist differences in POD activity in August and October between insect resistant and control Masson pine clones;2.74 compouds were detected from the volatile components of secondary metabolites,45 of which were identitied.There were 7 monoterpenes,22 sesquiterpenes,7 alcohols,3 esters, terpene oxidatives et al. The main compounds of masson pine needle were monoterpenes,sesquiterpenes and alcohols. Contents of volatile metabolites differed among months and among clones.There detected 16 different compounds between resistant clone and non-resistant clone.3.The contents of volatile metabolites in two insect resistant clones were different.In May,the needle monoterpene content of GK001 was significantly higher than those of GK002; In May,content of 9 compounds,which were Hexanal,2-Hexenal,a-Pinene,β-Myrcene,3,5-dimethyl-Octane,Sabinene,β-Ocimene,Terpinolene,Acetic acid,1,7,7-trimethyl-bicyclo[2.2.1]hept-2-yl ester,were significantly different between to insect-resistant clones.In October,the contents of 2 compounds,namely β-Ocimene,Ylangene,were significantly different between to insect-resistant clones.This study focused on differences of insect-resistant substances among different months and among different clones.This study might be benefitial to the research on insect-resistance mechanism of Pinus massoniana and breeding of insect-resistant masson pine.
