Botryosphaeria dothidea has become main plant disease of Carya cathayensis in the producingareas. This disease made a serious threat to industry, and made the growers to has a more economiclosses in recently. Now, there has no systematic research to detection and prevention, and can not testdirectly while the B. dothidea in the trees. So this thesis use the B.dothidea from Zhejiang Lin`an toidentify the molecular sequence of B.dothidea, research the biological characteristics, and test thesensitive of chemicals. As the result of establishment of the pathogen detection and prevention system.1.Identify system by molecularPrimers named tubulinF and tubulinR were designed. The sequences: tubulinF/tubulinR. B.dothidea has been made a PCR, It is a306bp to be get while other bacterial strain can notbeing got. The experiments show that the primers are specific. This primer can be used in all B.dothidea while all can be expanded.2.The research of biological characteristics.B.dothidea are often susceptible to environmental factors,for example, temperature, nitr-ogen,carbon,osmotic and pressure. The optimum temperature of B. dothidea is25℃.There is a maxof spore germination when the temperature is25℃. And when the temperature is15℃~30℃,The sugar、yeast powder of B.dothidea are the best carbon source and nitrogen source. The best pH of B.dothidea is6,and the best pressure of B.dothidea is when NaCl has0-0.1g per100mL.3.Control measures by chemical.Statistics found that the spore will be released at the early April to the end of July. The number ofthe spore is the top around the may every year. So we must prepare to preventing it before the April. Thesurvey found that trifloxystrobin is inappropriately to be used as the high price and the poor function inprevention. It is not appropriate to be used in disease control.
