Tomato yellow leaf curl disease (TYLCD) caused by Begomovirus is the most detrimentalfactor for tomato production all over the world. TYLCD occurred in greenhouse inthe Turpan District of Xinjiang in2013. The purposes of this research were to determine thevirus species of infected tomato plants, the biotype of invasive whiteflies and the incidence ofviruliferous whiteflies occurring in greenhouse, and to forecast the risk of TYLCD epidemics.The total of43tomato plants with yellow leaf curl symptoms collected from greenhousewere subjected to PCR-based diagnosis and DNA sequencing. The results showed that tomatoplants with viral symptoms were caused by Tomato yellow leaf curl virus (TYLCV), theincidence of diseased plant was39.5%.180whiteflies (Bemisia tabaci) were collected fromgreenhouse in Shanshan to identify their biotype and to investigate their capacity oftransmitting TYLCV. The biotypes were identified by PCR with primers specific for Qbiotype and B biotype of B. tabaci. Among the180whitefly samples,121were Q biotype,21were B biotype. The infection frequency of TYLCV in B. tabaci populations was investigatedby PCR using primers specific for TYLCV. In Q biotype whitefly, TYLCV were detectedfrom82, viruliferous percent was67.8%. In B biotype whitefly, TYLCV were detected from3,viruliferous percent was14.3%. The results showed that TYLCV were mainly transmitted byQ biotype whiteflies. A duplex PCR method was developed for the rapid detection of Qbiotype of whiteflies and the status of carrying TYLCV.The total of23Solanaceae vegetables varieties grown in Xinjiang were inoculated byinfectious clone of TYLCV. The results showed that TYLCV infect indigenous varieties oftomato, pepper and eggplant grown in Kashgar and showed yellow leaf curl symptoms. ThePCR detection results indicated that TYLCV can successfully replicated in plants. Itconcluded that TYLCV has potential risk of prevalent in Xinjiang.
