This studying was based on the materials of black spot pear, and separating and depuratingthe “Black spot disease” from it, then inoculated with pathogenic bacteria into pears, the majorpathogen was identified by morphological characters and internal transcribed spacer(ITS)analysis;Using different methods to deal with kurla pear, the pathogenic bacteria was inoculated back intogood pear, found the effect of different processing methods on it.Studyed on physiological andbiochemical indexes and disease resistance,to provide theoretical basis for the research, theoccurrence rules of kurla pear black spot, disease resistance and prevention and control measures.Then main results were as follows:1Isolating and Identifing of the Pathogen: It was classified in the Alternaria alternate (Fr.) Keissl.2Through the study of disease spot diameter, disease spot area, it was14mg/10L Ca>17mg/10LCa>23mg/10L Ca>20mg/10L Ca>11mg/10L Ca,0.5mg/kg1-MCP>0.3mg/kg1-MCP>0.2mg/kg1-MCP>0.4mg/kg1-MCP>0.1mg/kg1-MCP,200mmol/L Oxalic acid>25mmol/L Oxalicacid>400mmol/L Oxalic acid>100mmol/L Oxalic acid>50mmol/L Oxalic acid after storaged9days. Through the study on the depth of disease spot, it was14mg/10L Ca>17mg/10LCa>23mg/10L Ca>11mg/10L Ca>20mg/10L Ca,0.5mg/kg1-MCP>0.3mg/kg1-MCP>0.4mg/kg1-MCP>0.2mg/kg1-MCP>0.1mg/kg1-MCP,200mmol/L Oxalic acid>25mmol/L Oxalic acid>400mmol/L Oxalic acid>100mmol/L Oxalic acid>50mmol/L Oxalic acid after storaged9days.Removal of various processing of salicylic acid treatment, UV treatment, and other threetreatments selected three concentrations on physiological and biochemical research and diseaseresistance.3All of the1-MCP group’s treatment could improve the activity of SOD, activity of POD,activity of PAL of the late storage, the activity of the chitinase of the early storage and the latestorage, and reduce the cell membrane permeability of the middle storage.0.1mg/kg1-MCP could improve the activity of the early storage and the soluble protein content of the late storage.0.2mg/kg1-MCP and0.4mg/kg1-MCP could reduce the MDA content of the late storage. All ofthe Ca groups could keep the Vc content and the soluble protein content of the early storage;improve the activity of the late storage. All of the Ca groups could improve the chitinase of thestorage except the7days and21days, but the MDA content was higher than that of CK in thewhole storage. Although Ca treatment maintained the cell shape, but the choice of the cellmembrane permeability damaged, material inside and outside the cell membrane and leakage.20mg/10L Ca could improve the activity of PPO, the activity of the POD, the activity of the PAL.All of the oxalic acid groups could improve the activity of the PPO of the early storage, theactivity of the SOD, the activity of the POD, the activity of PAL, the activity of chitinase of thelate storage. In addition to the400mmol/L oxalic acid could keep the Vc content reduce the MDAcontent of the late storage.100mmol/L Oxalic acid the resistance of the various enzyme of the latestorage increase oxalate effect was more apparent,200mmol/L Oxalic acid and400mmol/L Oxalicacid could reduce the cell membrane permeability of the late storage.4Through the study of different means to deal with after inoculation of pathogenic bacteria offragrant pear research showed that: the disease spot area of oxalate inhibition effect of0.1mg/kg1-MCP,0.4mg/kg1-MCP,23mg/10L Ca,50mmol/L Oxalic acid,100mmol/L Oxalic acid,400mmol/L Oxalic acid is remarkable; all the processing method could restrain the extension ofdisease spot depth,0.1mg/kg1-MCP,100mmol/L Oxalic acid had the most obvious inhibitoryeffect on it. Each group could improve the activity of PPO of the early storage, the activity ofSOD of the late of the storage, the activity of the POD of the late storage, the activity of the PALof the late of the late storage, the activity of chitinase of the late storage;0.1mg/kg1-MCP has thebiggest effect on the activity of PPO,0.2mg/kg1-MCP has the biggest effect on the activity ofchitinase,0.4mg/kg1-MCP has the biggest effect on the activity of PAL. All of the Ca groupscould keep the Vc content of the late storage, inhibited the increase of MDA content, improved thecontent of soluble protein, the activity of the POD, the activity of PAL, the activity of chitinase.11mg/10L Ca has the obvious effect on increaseing the activity of PPO, inhibitong the rise of MDA;20mg/10L Ca has the obvious effect on the improving of the activity of SOD, increasingthe soluble content, increase the activity of chitinase;23mg/10L Ca has the obvious effect onreduce the cell membrane permeability, enhance the activity of PAL. All of the oxalic acid group’streatment could inhibit the increasing of the content of MDA, increasing the activity of PPO,increasing the activity of POD, increasing the activity of PAL, and increasing the content ofsoluble protein.100mmol/L Oxalic acid has the most obvious effect on it;50mmol/L oxalic acidhas the obvious effect on increasing the activity of POD, reduced the cell membrane permeability,increased the soluble protein.5By room temperature storage and damage inoculation of fragrant pear each measurement indexof the contrast: By ambient storage pear after inoculation of pathogenic bacteria and compareresults was0.4mg/kg1-MCP>20mg/L Ca>100mmol/L Oxalic acid, therefore0.4mg/kg1-MCPcould delay the fruit postharvest metabolic rate, enhance the resistance of most of the enzymesactivity, which was beneficial to strengthen the disease resistance to black spot of fragrant pear.
