Equine rhinopneumonitis (ER) is a collective term for any one of several highly contagious,clinicaldisease entities of equids that may occur as a result of infection by either of herpesviruses,Whichcharacterized by a primary respiratory tract disease of varying severity.Infections by EHV-1in particularare capable of progression beyond the respiratory mucosa to cause the more serious disease manifestationsof abortion, perinatal foal horse death,or EHM.According to published EHV-1gB and gD sequence planed two pairs of primers to amplify the targetgenes,then tandem clone genes to the vector pET-30a.And transformed them to E.coli BL21.SDS-PAGEanalysis showed that the expressed gB and gB-gD was26kD and50kD in molecular massrespectively.Western blotting analysis showed that the two proteins could react with equine herpesvirustype1serum and had a good reactivity.The healthy mice were immunized with gB、gD and gB-gD protein,the levels of anti-protein IgG andthe proliferative reaction of lymphocytes were detected by ELISA and MTT.The dates showed thatcontrasted with the juxtaposed group,both of the protein groups produced a significant difference and thefusion group is better(P<0.01).To know the prevalence of ER in Xinjiang,a ER sero epidemiological investigation was performed in5counties(cities)of Xinjiang with3195Equines for the years2012and2013and texted by indirectELISA.Result show that the positive serological reaction is1237cases(38.71%).Among them,the highestpositive rate season is Autumn(45.85%),followed by Summer(43.44%),the lowest is Winter(19.42%).AndER’s positive rates in2013were significantly lower than2012(P<0.01).Based on generous census data toprovide reference for equine rhinopneumonitis prevention work and the healthy development of Xinjianghorse industry.In summary,the study laid the foundations for the monitor of ER in Xinjiang and the development ofsubunit vaccine in the future.
