Epidemiological Investigation In Stud-farms In 2 Counties Of Xinjiang And Comparative Study On Two Indirect ELISA Detection Methods Of Equine Rhinopneumonitis

This research aimed at infection condition investigation of equine rhinopneumonitis, equine influenza virus, equine salmonella and strangles in stud-farms in Zhaosu County and Fuyun County, Xinjiang and compared specificity, sensitivity and repeatability of the indirect ELISA detection method for equine rhinopneumonitis constructed in our laboratory and a commercially available indirect ELISA detection kit. 502 serums were sampled in stud-farms in Zhaosu County(227 samples) and Fuyun County(225 samples), Xinjiang in this experiment. Equine rhinopneumonitis, equine salmonella and strangles in these serums were tested with an indirect ELISA method. Equine influenza virus in these serums was tested by hemagglutination inhibition tests. Equine rhinopneumonitis antibodies in 227 samples of equinum serums of Zhaosu County, Xinjiang were tested by the indirect ELISA detection method of equine rhinopneumonitis constructed in our laboratory and a commercially available indirect ELISA detection kit respectively and their outcomes were compared. Results showed: 1. Epidemiological investigation of equine rhinopneumonitis and equine influenza virus: the average positive rate of equine rhinopneumonitis antibodies was 15.16% in Zhaosu County and 12.00% in Fuyun County; the average positive rate of equine influenza antibodies was 24.19% in Zhaosu County and 16.00% in Fuyun County; the overall positive rate of antibodies of both equine rhinopneumonitis and equine influenza was 3.61% in Zhaosu County and 4.89% in Fuyun County. 2. Epidemiological investigation of Streptococcus equi and equine salmonella: the average positive rate of Streptococcus equi antibodies was 20.94% in Zhaosu County and 8.44% in Fuyun County; the average positive rate of equine salmonella antibodies was 26.71% in Zhaosu County and 6.22% in Fuyun County; the overall positive rate of antibodies of both Streptococcus equi and equine salmonella was 7.94% in Zhaosu County and 3.11% in Fuyun County. 3. 227 samples of equinum serums of Zhaosu County, Xinjiang were tested by the indirect ELISA detection method of equine rhinopneumonitis constructed in our laboratory and a commercially available indirect ELISA detection kit respectively. The results of self-constructed ELISA detection showed that there were 42 positive samples and 235 negative samples; the results of the commercially available kit showed that there were 39 positive samples and 238 negative samples. A commercially available kit was compared with testing results of self-constructed ELISA detection and the positive coincidence rate, the negative coincidence rate and the overall coincidence rate were 91.36%, 97.48% and 97.11% respectively.Epidemiological investigation of horse infectious diseases in stud-farms in 2 counties of Xinjiang provided reliable reference data for prevention and control of horse infectious diseases in Xinjiang. The indirect ELISA detection method for equine rhinopneumonitis constructed in our laboratory was compared with a commercially available indirect ELISA detection kit with an overall coincidence rate of 97.11%, confirming that specificity, sensitivity and repeatability of the indirect ELISA detection method constructed in our laboratory met the commercial standard. It provided data support for the indirect ELISA detection method of equine rhinopneumonitis constructed in our laboratory.
