Patinopecten yessoensis is a cold-water shellfish,the individual is large,and fast growth, is an important marine food economy shellfish of our country,also,it is have large medicinal and health effects.Patinopecten yessoensis is introduced from Japan in the1980s, now become one of the main Northern seawater shellfish.The scraps were produced in the process of processing Patinopecten yessoensis,these scraps did not get the full development and utilization of this in-depth study of Patinopecten yessoensis processing waste glycosaminoglycan can Patinopecten yessoensis waste provide a theoretical basis for development of high-value resources, explore a new way for the development of new drug lead compounds.The research of Patinopecten yessoensis scraps glycosaminoglycan was investigating the glycosaminoglycan’s separation and purification and the biological activities.The main content and the results are as follows:1, Based on single factor experiments, we use of the RSM optimizating the extraction process of glycosaminoglycan extraction.Experiment quadratic regression equation to determine the optimum extraction conditions of glycosaminoglycan enzymatic pH value of8.00, reaction temperature40.00℃, hydrolysis time3.5h, solid-liquid ratio of1:2, in this condition lower extraction rate reach0.265%.2, Measuring the chemical composition of Patinopecten yessoensis scraps glycosaminoglycan,the results show that the sulfate glycosaminoglycan content of5.75%±0.42%,25.86%±1.35%uronic acid content, amino sugar content1.03%±0.26%.Using infrared spectroscopy analysis showed that the functional groups of the glycosaminoglycan-OH (3424cm-1),containing the shrimp scallop scraps glycosaminoglycan-COO-(1653cm-1)and-NH(1567can-1),-SO2(1414cm-1), C-O-C(sugar ring,1044cm-1) and other characteristics group.3, Research the activity of Patinopecten yessoensis scraps glycosaminoglycan biological.The results showed that the glycosaminoglycan has good scavenging hydroxyl radicals, superoxide anion radical and DPPH radical capacity and reducing power,better ability to chelate metal;glycosaminoglycan hepatoma cell line HepG2cells strong inhibition;vitro experiments confirmed that glycosaminoglycans may be extended TT, PT, APTT,but compared with heparin activity was low.
