Tripterygium wilfordii Hook, as a Chinese medicinal crop, is wide distributed in Fujian province. It is mainly used for the therapy of inflammation, cancer and chronic nephritis etc. Some monomer components, such as Triptolide and Tripterine, had been well developed and used for clinical. This study used T. wilfordii which is wide cultivated in Taining of Fujian province as material, the methods for detection and analysis of Triptolide and alkaloid, the difference of the content of Triptolide and total alkaloids with different growth years, collecting seasons and different collecting parts were the key researchs. The harvesting standards of the leaves and roots of T. wilfordii were formulated, main results are as follows:1. The methods of absolute ethyl alcohol for extraction, column chromatography for decontamination, HPLC for the detection of the content of Triptolide was established. Also, the methods of absolute ethyl alcohol for extraction, column chromatography for decontamination, uv-vis spectrophotometry for the detection of the content of total alkaloids was etablished. And the results showed that the methods had good repeatability and it’s results were reliable.2. The methods of absolute ethyl alcohol for extraction, column chromatography for decontamination, HPLC for detection to analyze the different contents of Triptolide with different growth years, collecting seasons and different collecting parts was used. And the results indicated:①growth years had effects to the contents of Triptolide in the leaves and roots of T. wilfordii, the leaves of T. wilfordii of six years old had the maximum content of Triptolide(136.33±6.19μg·g-1), the roots of T. wilfordii of five years old had the maximum content of Triptolide(118.33±5.71μg·g-1);②the content of Triptolide in the leaves and roots of T. wilfordii were different with different collecting seasons, the leaves of T. wilfordii collected in august had the maximum content of Triptolide(131.73±8.91μg·g-1), the roots of T. wilfordii collected in october had the maximum content of Triptolide(132.33±7.21μg·g-1). 3. The methods of absolute ethyl alcohol for extraction, column chromatography for decontamination, uv-vis spectrophotometry for detection to analyze the different contents of total alkaloids with different growth years, collecting seasons and different collecting parts was used. And the results indicated:①growth years had effects to the contents of total alkaloids in the leaves and roots of T. wilfordii, the leaves of T. wilfordii of six years old had the maxlmum content of total alkaloids(238.47±10.38μg·g-1), the roots of T. wilfordii of five years old had the maxlmum content of total alkaloids(622.38±12.43μg·g-1);②the content of total alkaloids in the leaves and roots of T. wilfordii were different with different collecting seasons, the leaves of T. wilfordii collected in august had the maxlmum content of total alkaloids (223.19±12.38μg·g-1), the roots of T. wilfordii collected in October had the maxlmum content of total alkaloids (630.43±11.59μg·g-1).4. The harvesting standards of T. wilfordii in the producing area of Taining (Fujian province) were formulated, and the standards were very suitable for the extraction of Triptolide.
