Porcine reproductive and respiratory syndrome (PRRS), commonly known asblue ear disease,Porcine reproductive and respiratory syndrome virus (PRRSV)causes respiratory diseases, the failure of sows and piglets. The virus ishighly-contagious making the body immune suppression and mixed infection. Porcinereproductive and respiratory syndrome virus caused huge losses, the development ofanimal husbandry.Usually after7~10d PRRSV infected, swines will appear humoral immune ofantigen specificity, and it can be detected neutralizing antibody in the serum from14d to28d.Interleukin12(IL-12) is a kind of important white blood cells, immune,which is multifunctional, Involved in nonspecific and specific immunity of early.This study regard swines immuned with PRRS as our subject investigated,analysising the secretion of IL-12under the stimulation of specific peptide ofPRRSV,and then understanding the characteristics of which lymphocyte in peripheralblood secrete IL-12. To find out reasonable clinical immune program. This studyestablishes the swine model with PRRS immuned, Through the determination ofELISA of porcine peripheral blood lymphocyte interleukin, stimulation in peripheralblood lymphocytes is performed using PRRSV specific peptide. Our previousexperiments have shown that the pig immune PRRSV attenuated vaccine or PRRSVpeptide stimulation, after a period of time the body IL-12content will be increased,enhance the immunity of pigs.The results shows that the level of IL-12stimulated with PRRSV-GP5aa100-108and PRRSV-GP5aa96-104100-108less than the control group. Porcinereproductive and respiratory syndrome virus immune, IL-12synthesis of extendedlevel has increased, but the total trend. It showed that PRRSV inhibited the GP5secretion of IL lymphocytes in peripheral blood of12.The level of IL-12in peripheral blood stimulated with PRRSV-N106-114at different time points has been decreased in high antibody group and increased in lowantibody group after PRRS immune14d. It shows that in the early vaccination withPRRSV when the vaccinated well PRRSV-N106-114inhibited the secretion of IL-12if not will promote the secretion of IL-12. And after PRRS immune28d thelymphocytes stimulated with PRRSV-N106-114, The secretion of IL-12increased inhigh antibody group and decreased in low antibody group. It shows that when thevaccinated well the level of the antibody content is increased. The PRRSV-N106-114will promote the peripheral blood lymphocytes secrete the IL-12.If not will inhibit thethe secretion of IL-12.The level of IL-12in peripheral blood stimulated with PRRSV-N49-57atdifferent time points in high antibody group are promoted in the early then inhibitedand in low antibody group the difference is not obvious after PRRS immune14d. Itshows that in the early PRRS immune PRRSV-N49-57will inhibit the secretion ofIL-12. And at28d the secretion of IL-12are promoted in the early then inhibited inhigh antibody group and it is inhibit in low antibody group. So we can conclude thatwhen vaccinate with PRRSV well PRRSV-N49-57could promote the secretion ofIL-12in the early then inhibit it and if not it will inhibit the secretion of IL-12.These results indicate that, vaccinated pigs depending on the secretion of IL-12.The study of PRRSV specific peptide is very useful for the development of the PRRSvaccine and infection of PRRSV. The degree of immune is discrepant with differenceof immune times, immune periods, specific peptides and immune programs. IL-12hasbidirectional regulation effect on PRRS immunity.The relationship between IL-12andcell function and how the PRRSV specific peptide affects the immune mechanismwill study further.
