Effects Of Thymopentin On Production Performance And Antioxidant Function In AA Broilers

Abstract/Summary:
This research were aimed to investigate the infection on production performance, blood physiological and biochemical indicators, morphological characteristics of immune organs, change of antioxidant enzyme activity and free radical in serum, liver,spleen and immune organs in broilers via intramuscular thymopentin injection. And discuss the mechanism of antioxidation of thymopentin promote the immunity, then provide test and theoretical basis for the usage in herds’ productivity.Methods:1) To examine the effect of thymopentin on the blood physiological and biochemical indicators, antioxidant function of serum, liver, kidney, spleen, thymus and bursal tissue, morphological characteristics of immune organs in broilers,15014-day-old healthy avian broilers were randomly divided into five groups by gender and intramuscular thymopentin (TP-5) at a dosage of50,150,300,500μg·kg-1, whereas the control group weight. And four treatment groups (TG)(group Ⅰ, Ⅱ,Ⅲ and Ⅳ) were injected separately with (CG)(group V) was given the same dosage by the aseptic physiological saline method.2) Before the test, the blood physiological and biochemical indicators was investigated by using tube dilution method and AUTOLAB automatic biochemical analyzer of AMS made in Italy. Results were also obtained on Day6and Day10of the research.3) For five days, the daily change of the antioxidant indices of blood, liver and kidney in broilers was investigated. Results were also obtained on Day10of the research. The Day6(19-day-old) and Day10(24-day-old) results were then compared using ELISA kits.4) For five days, the daily change of the antioxidant indices of spleen, thymus and bursal in broilers was investigated. Results were also obtained on Day10of the research. The Day6(19-day-old) and Day10(24-day-old) results were then compared using ELISA kits.5) Pathological lesions of organs in broilers were observed.Results:1) The infection on production performance:During the treatment (between14-day-old to19-day-old) results for TG vs CG, showed that ADG of group Ⅰ,Ⅱ increased (P<0.05或P<0.01). F/G of group Ⅰ, Ⅱ decreased (P<0.01). After the treatment (between19-day-old to24-day-old) results for TG vs CG. showed that ADG of group Ⅰ. Ⅱ decreased (P<0.05或P<0.01), F/G of group Ⅰ, Ⅱ increased (P<0.01).2) The effect of thymopentin on the blood physiological and biochemical indicators:Results for TG vs CG, showed that in the TG, lymphocyte percentage of group Ⅰ, Ⅱ, IV increased (P<0.05), the CRE、AST、GGT contents in serum decreased (P<0.05), the ALT contents in serum increased (P<0.05).3) The effect of thymopentin on the antioxidant function of serum, liver and kidney tissue in broilers:The Day6results for TG vs CG, showed that in the TG, the CAT contents in serum and liver of group Ⅱ increased (P<0.05or P<0.01), the SOD, GSH-Px contents in serum and liver of group Ⅰ,Ⅱ increased (P<0.05), the SOD contents in kidney and the T-AOC in serum of TG increased (P<0.05), the MDA contents in serum, liver and kidney of group Ⅰ,Ⅱ decreased (P<0.05), the T-AOC in liver of group Ⅰ,Ⅱ and in kidney of group Ⅱ decreased (P<0.05). The Day10results for TG vs CG, showed that the SOD, GSH-Px and CAT contents and the T-AOC in serum, liver and kidney increased as before (P<0.05), the contents of oxygen free radicals in broilers increased with age and weight, the GSH-Px contents in serum and kidney of TG decreased (P<0.05), the T-AOC in serum and liver of TG decreased (P<0.05).4) The effect of thymopentin on the antioxidant function of immune organs in broilers:The Day6results for TG vs CG, showed that in the TG, the CAT contents in spleen of group Ⅰ, Ⅱ and in thymus of group Ⅱ increased (P<0.05or P<0.01); the GSH-Px contents in spleen, thymus and bursal of group Ⅰ and Ⅱ increased (P<0.05); the SOD contents in spleen and thymus of group Ⅰ,Ⅱ increased (P<0.05or P<0.01); the MDA contents in spleen of group Ⅱ and thymus of group Ⅰ,Ⅱ decreased (P<0.05); the T-AOC in immune organs of group Ⅰ, Ⅱ and in thymus of group Ⅲ, Ⅳ increased (P<0.05). The Day10results for TG vs CG, showed that the SOD,GSH-Px and CAT contents and the T-AOC in immune organs of group Ⅱ increased in TG as before (P<0.05); the contents of oxygen free radicals in broilers increased with age and weight, the GSH-Px contents in spleen and the CAT,GSH-Px contents in thymus of TG decreased (P<0.05), the SOD contents in thymus of TG increased slightly (P>0.05), the T-AOC in bursal of TG decreased (P<0.01).5) The effect of thymopentin on the immune organs in broilers:The Day6results for TG vs CG, showed that in the TG, Wet and dry weight ratio of Immune organs of group~,Ⅱ increased (P<0.05), the difference between group Ⅲ,Ⅳ and CG was not significant (P>0.05). The Day10results for TG vs CG, showed that Wet and dry weight ratio of Immune organs of group Ⅰ, Ⅱ in TG increased as before (P<0.05). In the group at dosage500μg·kg-1, there were minimal splenorrhagia and splenaemia, partial cell split, and the cells of thymus and bursa fabricius were uniform, clean, and clear, non-pathologic histology changes.Conclusion:The results showed that TP-5could improve the production performance of broiler during the treatment. TP-5could improve the activity of the antioxidant enzyme and reduce the level of oxygen free radicals. The optimum injection dosage was found to be50,150μg· kg-1. There were no obvious damage to immune organs, liver and kidney tissues and non-pathologic histology changes.
