Cryptomehia fortunei is an important high altitude afforestation tree species in south of China.In order to enrich and Preserve germplasm resources, we collectted49clones and evaluated thediversity of C. fortunei resources. The results of this study are summarized as follows:1. With24299ESTs sequence of Cryptomeria japonica,70EST-SSR markers were exploid,18pairs of which are polymorphic markers.2. EST-SSR and SSCP markers were used to analysis the genetic diversity of C. fortunei resources,The results showed that the shannon’s information index(I)、observed number of alleles(Na)、effective number of alleles(Ne) and Nei’s gene diversity(H) were0.5517、1.9379、1.6746、0.3789,respectively. Based on molecular markers, dendrogram were constructed. The results showed that49different materials were divided into3groups at coefficient rate of0.56with the first group wereFujian Laizhou resources and the second group are mixed resources, and the third group are FujianLayang resources. The genetic relationships of Fujian C. fortunei resources were relativelyindependent. However, Zhejiang resources were relatively close relationship.3. From initial49C. fortunei resources,23optimal clones were chosen for hybridization parents.Results of t-test showed that the observed number of alleles, effective number of alleles, Nei genediversity index had no significant differences between optimal hybridization parents group and theinitial germplasm on0.01level. These results showed that the optimal parents can represent initialgermplasm resources very well.4. Ten cross combination were made, in which2clones of C. fortunei were chosen from23optimalparents groups as male parent and5excellent clones were chosen from Laoshan forest farm asfemale parent. The results showed that the cones developed well.
